摘 要:目的获得FACSCount计数仪测量的我国部分省市和民族健康青少年及成年人群外周血T淋巴细胞亚群绝对数参考值范围。方法从吉林、河南驻马店、山西、贵州、四川凉山州和云南德宏州6省市当地体检中心常规体检个体(常规体检血液)中获得符合纳入标准的样本1 276份,使用FACSCount计数仪检测EDTA抗凝全血样本中CD3+、CD4+、CD8+淋巴细胞的绝对数并计算CD4+/CD8+绝对数比值。正常参考值范围用中位数和各参数值分布曲线95%的曲线下面积表示。结果 CD3+、CD4+、CD8+淋巴细胞的绝对数中位数分别为1 472、768、564个/μl,CD4+/CD8+绝对数比值中位数为1.42。年龄、民族为其影响因素,CD3+、CD8+淋巴细胞绝对数与年龄呈负相关,Spearman等级相关系数分别为-0.12、-0.17;CD4+/CD8+绝对数比值与年龄呈正相关,Spearman等级相关系数0.19,P<0.01。各参数值在不同性别间分布差异无统计学意义。CD3+、CD4+淋巴细胞绝对数汉族为4个民族中最低;CD8+淋巴细胞绝对数彝族最高,CD4+/CD8+绝对数比值傣族为4个民族中最高。结论在本研究中,各参数在不同年龄组和不同民族间分布差异有统计学意义,年龄、民族为其影响因素。Objective To establish normal reference of peripheral blood lymphocyte subsets in healthy teen- agers and adults. Methods A total of 1 276 whole blood samples were collected from healthy individuals for regular physical examination in local medical centers of Jilin, Henan, Shanxi, Guizhou, Sichuan and Yunnan provinces of China. The absolute number of CD3+ , CD4+ , CD8+ T-lymphocytes in EDTA anti-coagulant blood samples were count by FACSCount cytometry. The reference value was determined by the 2.5 and 97.5 percentiles of the parameters. Results The median values of the T lymphocyte subsets were 1 472 cells//A for CD3+, 768 cells/μl for CD4+, 564 cells/μl for CD8+ and a ratio of 1.42 for CD4+/CD8+. The results showed significant differences of each parameter at different age and ethnic groups, but no significant differ- ence was observed in gender. Age-related decrease was observed in CD3+ and CD8+ cell counts with the Spearman's correlation coefficients at - 0.12 and - 0.17, respectively. Age-related increase of the CD4 + / CD8+ ratio was observed with the Spearman's correlation coefficients at 0.19 (P〈0.01). Conclusions Nor- mal reference values of T-lymphocyte subsets isolated from healthy individuals in 6 provinces of China were es- tablished and the results showed that age and ethnic groups had an impact on the lymphocyte subset levels in the peripheral blood.
