摘  要:目的通过对比分析正常人和高尿酸血症患者血清蛋白双向电泳图谱,初步分析高尿酸血症血清差异蛋白。方法采用双向凝胶电泳(2-dimentional gel electrophoresis,2-DE)和基质辅助激光解吸飞行时间质谱(matrix-assisted laser desorption/ionization time-of-flight tandem mass spectrometry,MALDI-TOF-MS)对高尿酸血症患者和对照组健康人血清进行分析,寻找高尿酸血症血清差异蛋白。结果差异表达蛋白质点数为10个,质谱成功鉴定出4个差异蛋白质,其中补体C3、触珠蛋白、α1-抗胰蛋白酶呈上调表达,载脂蛋白L1呈下调表达。结论补体C3、触珠蛋白、α1-抗胰蛋白酶、载脂蛋白L1在高尿酸血症组和正常对照组血清中存在差异,但结果有待运用其他生物学的方法进一步验证。Objective To study the differentially expressed serum proteins between controls and patients with primary hyperuricemia (HUA) by two-dimensional gel electrophoresis (2-DE). Methods 2-DE and matrix-assisted laser desorption/ ionization time-of-flight tandem mass spectrometry( MALDI-TOF-MS ) were used to detect the serum of controls and patients with HUA for screening the differentially expressed serum proteins. Results There were 10 differentially expressed serum protein spots by gel-image analysis. Four differentially expressed serum proteins were successfully identified with MALDI-TOF-MS. The complement 3, haptoglobin and otl-antitrypsin were up-regulated and apolipoprotein L1 was down-regulated in patients with HUA, as compared with controls. Conclusion The complement 3, haptoglobin, α1-antitrypsin and apolipoprotein L1 are differentially expressed between controls and patients with HUA and the result should be validated by other biochemical technologies.
