摘  要:目的探讨油酸对3T3-L1脂肪细胞脂联素表达的影响及PPARγ介导的机制。方法不同浓度油酸处理体外培养成熟的3T3-L1脂肪细胞24h,并选取100μmol/L油酸加与不加过氧化物酶体增殖物激活受体γ(PPARγ)拮抗剂GW 9662处理3T3-L1脂肪细胞,运用实时荧光定量PCR方法检测处理前后脂联素及PPARγmRNA的表达水平,并用western-blotting法测定脂联素蛋白表达水平。结果与对照组相比,油酸在浓度为25、50、100μmol/L时上调脂联素及PPARγmRNA的表达,随着浓度的增加脂联素和PPARγmRNA的表达下降;油酸中加入GW 9662处理后脂联素mRNA及蛋白的表达水平分别降低77%、78.01%(P<0.05)。结论油酸在一定浓度范围内能上调3T3-L1脂肪细胞脂联素及PPARγ基因表达,且呈剂量依赖关系,加入PPARγ拮抗剂GW9662后能够阻断这种上调作用,提示油酸可通过激活PPARγ来调节脂肪细胞脂联素的表达。Abstract: Objective To explore the effects of oleic acid on the gene and protein expression of adiponectin and its PPARγ mechanism in 3T3-11 adipocytes. Method 3T3- L1 adipocytes were incubated with different concentration ( 25,50,100,200,400 μmol/L) for 24h, and the mRNA expression of adiponectin and PPART was determined by real-time PCR (relative quantity). And then the 3T3-L1 adipocytes were treated for OA at concentration of 100μmol/L for 24h with/without the affect of PPARγinhibitor GW9662, to determine the gene and protein expression of adiponectin. The protein expression changes of adiponectin were detected by immunoblotting. Result The mRNA expression of adiponectin and PPARγ increased at the dosages of 25,50, and 100μmol /L. With the increase of OA, the mRNA expression of adiponectin and PPART decreased in a dose- dependent manner. Incubating with GW9662 and OA, decreases in the mRNA and protein expression of adiponectin were 77% and 78.01% ( P 〈 0. 05 ), compared with the control respectively. Conclusion OA could increase the gene expression of adiponectin andPPARγ mRNA in dose-dependent manner in 3T3-L1 adipocytes,which could be specifically blocked by GW9662,OA indues the expression of adiponectin mRNA and protein in 3T3-L1 adipocytes, possibly by a mechanism involving PPARγ.
