复方胃肠安对人大肠癌裸鼠原位移植瘤生长和肝转移的影响

摘　　要：目的观察以健脾类中药为基础的复方胃肠安对人大肠癌裸鼠原位肿瘤生长和肝转移的抑制作用。方法采用人大肠癌HCT116细胞制作原位移植瘤模型,将造模后的裸鼠随机分为对照组、胃肠安组、5-Fu组和胃肠安+5-Fu组。各组采取相应的干预措施。观察各组原位肿瘤的质量、体积、抑瘤率,各组的肿瘤转移特别是肝转移情况,各组原位肿瘤和肝转移瘤的病理形态,以及血清癌胚抗原(CEA)、甲胎蛋白(AFP)浓度。结果各组裸鼠原位移植成瘤率为100%。5-Fu组和胃肠安+5-Fu组的原位肿瘤平均质量均显著低于对照组(P<0.01),胃肠安+5-Fu组的原位肿瘤平均体积显著小于对照组(P<0.05),胃肠安组、5-Fu组和胃肠安+5-Fu组的抑瘤率分别为19.087%、33.298%、36.802%。胃肠安组和胃肠安+5-Fu组的肝、腹壁、肠系膜等的转移明显少于对照组(P<0.05,P<0.01),对照组、胃肠安组、5-Fu组和胃肠安+5-Fu组的总转移率分别为100.00%、50.00%、72.72%、40.00%。胃肠安+5-Fu组的血清CEA水平显著低于对照组(P<0.05)。结论复方胃肠安对人大肠癌裸鼠原位肿瘤生长和肝转移具有一定的抑制作用,在配合化疗使用后效果较为显著。Objective To evaluate the inhibitory effect of ＂Weichang＇ an Decoction＂ on the growth and hepatic metastasis of orthotopic transplant in nude mouse models of human colorectal cancer. Methods The nude mice were implanted with HCT116 cells at orthotopic site, then randomly divided into four groups： control group, Weichang＇ an group, 5-Fu group, Weichang＇ an ＋ 5-Fu group. Each group Was treated with corresponding intervention. The mass, volume, growth- inhibiting rate of orthotopic tumors and the metastatic lesions, the pathomorphology of orthotopic tumors and hepatic metastasis, the concentrations of CEA and AFP were detected. Results The orthotopic tumor formation rate was 100% in each group. Compared with the control group, the mass （ P 〈 0.01 ） and volume （ P 〈 0.05 ） in 5-Fu group and Weichang＇ an ＋ 5-Fu group were decreased significantly. The growth-inhibiting rates of orthotopic tumors in Weichang＇ an group, 5-Fu group and Weichang＇ an ＋5-Fu group were 19. 087%, 33. 298% and 36. 802% respectively. The incidence rates of metastasis in liver, abdominal wall, mesentery et al in Weichang＇ an group （P 〈0.05 ） and Weichang＇ an ＋ 5-Fu group （P 〈 0. 01 ） were lower than the control group obviously; the total metastasis rates in control group, Weichang＇ an group, 5-Fu group, Weichang＇ an ＋5-Fu group were 100%, 50%, 72.72% and 40% respectively. The concentrations of CEA in Weichang＇ an ＋ 5-Fu group were lower than that of the control group significantly （ P 〈 0. 05 ）. Conclusion ＂Weichang＇ an Decoction＂ has certain inhibitory effect on the growth and hepatic metastasis of orthotopic transplant nude mouse model of human colorectal cancer. Combination of ＂Weichang＇ an Decoction＂ with chemotherapy shows better results.
