摘  要:目的探讨小檗碱(berberine,BBR)对糖尿病肾病(diabetic nephropathy,DN)大鼠肾病变的保护作用及其部分机制。方法以高脂饲料喂养联合低剂量链脲佐菌素(STZ)诱导DN大鼠模型,动物分为正常对照组、模型组、3个BBR治疗组、依那普利治疗组、消渴丸治疗组。给药8周后取大鼠肾脏,Masson染色观察肾组织形态学改变;免疫组织化学检测肾组织纤维连接蛋白(FN)与结缔组织生长因子(CTGF)的表达。结果与模型组比较,BBR可改善DN大鼠肾脏形态学异常,降低肾组织中FN与CTGF的表达。结论 BBR可延缓糖尿病大鼠肾脏损伤,此作用可能与下调肾脏组织FN与CTGF的表达有关。Objective To investigate the effect and mechanism of berberine on diabetic nephropathy rat model. Methods The rat model of diabetic nephropathy was induced by intraperitoneal injection of streptozoein ( STZ ) after having high-fat diet for six weeks. The rats were assigned into 7 groups:normal control group, model group,3 berberine treatment groups, diabetic rats treated with enalapril and Xi- aoKe Wan every day served as positive control groups. After treatment for 8 weeks, each left kidney was removed. Masson' s trichrome staining was used to observe the pathological changes of kidney. The expression of fibronectin ( FN ) and connective tissue growth factor (.CTGF) were detected by immunohistoehemistry. Results Berberine could suppress alterations in histological changes in the kidney tis- sues, decrease the expression of FN and CTGF in renal tissue of diabetic rats with nephropathy. Conclusion Berberine may slow down the progression of diabetic nephropathy in rats by suppressing the expression of FN and CTGF in renal tissue.
