摘 要:目的探讨白细胞介素17A(IL-17A)、核因子κB(NF-κB)、孕激素受体(PR)与自然流产的关系。方法随机选取30例早期自然流产患者作为流产组,30例正常早孕人工流产孕妇为对照组。酶联免疫吸附法(ELISA法)检测蜕膜组织中IL-17A、NF-κB与PR蛋白的表达;免疫组化法检测IL-17A、NF-κB与PR在蜕膜组织中的表达,对其进行定位、半定量分析,并对其之间的相互关系进行分析。结果①流产组蜕膜组织中IL-17A和NF-κB蛋白表达水平高于对照组,PR蛋白表达水平低于对照组,差异有统计学意义(P〈0.05)。②流产组胞质中IL-17A的表达高于对照组(P〈0.05);流产组核内NF-κB的表达高于对照组(P〈0.05),胞质中NF-κB的表达低于对照组(P〈0.05);流产组胞质中和核内PR的表达低于对照组(P〈0.05)。③胞质中IL-17A与胞核中NF-κB呈正相关(r=0.500,P〈0.05),与胞质中NF-κB呈负相关(r=-0.499,P〈0.05);核内NF-κB与核内PR呈负相关(r=-0.513,P〈0.05),核内NF-κB和与胞质内PR呈负相关(r=-0.511,P〈0.05)。结论蜕膜组织中IL-17A表达增强可能与不明原因早期自然流产有关。蜕膜组织中IL-17A的活化可增加NF-κB表达,进而减少PR表达并减弱其功能,可能是其发病机制之一。Abstract: Objective To evaluate the expression of interleukin 17A ( IL - 17A), nuclear factor κB ( NF - κB) and progesterone receptor (PR) in the decidual tissue of unexplained first trimester spontaneous abortion, and to analyze the mechanisms of the changes of IL - 17A, NF - κB and PR in the decidual leading to abortion. Methods 30 first tri- mester spontaneous abortion and 30 normal first trimester pregnant women were randomly devided as abortion group and control group. Expression of IL - 17A, NF - κB and PR protein in deciduas of abortion group and control group was de- teted by enzyme linked immunosorbent assay (ELISA). Immunoflunistochemistry was employed on abortion group and control group to show the location, quantity and interaction of IL - 17A, NF - κB and PR. Results ① Expression of proteion of IL- 17A, NF- κB was significantly higher in decidual of abortion group compared with that of control group ( P 〈 0. 05 ), and the PR proteion was significantly lower ( P 〈 0. 05 ).② The quantities of intracytoplasma IL - 17A are higher in abortion group than control group(P 〈0.05 ) ; Intranuclear NF - κB was higher in abortion group than control group (P 〈0.05), but intracytoplasma NF - κB was lower in abortion group than control group(P 〈0.05) ; Both the in- tracytoplasma PR and intranuclear PR were lower in study group than control group( P 〈 0. 05, P 〈 0. 05 ). ③Intracyto- plasma IL - 17A had a positive correlation with intranuclear NF - κB ( r =- 0. 500, P 〈 0.05 ) , and a negative correlation with intracytoplasma NF - κB (r = -0. 499, P 〈 0.05 ) ; intranuclear NF - κB had a negative correlation with intranuclear PR ( r = - 0. 513, P 〈 0.05 ), and a negative correlation with intracytoplasma PR ( r = - 0. 511, P 〈 0.05 ). Conclusion The upregulation of IL - 17A in decidual of abortion group may contribute to unexplained first trimester spontaneous a- bortion. The activation of IL - 17A may increase the expressi
