摘 要:目的:观察水飞蓟素对载脂蛋白E(ApoE)基因敲除(ApoE-/-)小鼠动脉粥样硬化的影响,并探讨其可能机制。方法:高脂喂养加缩窄性颈动脉套管术诱导小鼠动脉粥样硬化形成。实验分对照组(A组)、水飞蓟素低剂量组(B组)与水飞蓟素高剂量组(C组)。给药4周后取颈动脉标本,测量斑块面积、纤维帽厚度及脂质含量;检测血脂及血清丙二醛、超氧化物歧化酶水平。结果:①A、B两组间血脂水平差异无统计学意义,C组较A、B两组血脂均降低(P<0.05);②与A组相比,B、C两组斑块面积及脂质含量均减少(P<0.05),纤维帽厚度均增加(P<0.05),且C组较B组改变更显著(P<0.05);③与A组相比,B、C两组血清丙二醛水平均降低(P<0.05),超氧化物歧化酶水平均增高(P<0.05),且C组较B组改变更显著(P<0.05)。结论:水飞蓟素对ApoE-/-小鼠已形成的动脉粥样硬化斑块进展有明显抑制作用,其机制可能与降脂和抗氧化应激有关。Objective:To examine the effect of silymarin on atherosclerosis in apolipoprotein E-deficient(ApoE-/-) mice,and explore the potential mechanisms. Method:Carotid atherosclerotic lesions were induced by high-fat diet and perivascular constrictive collars placement.ApoE-/-mice were randomly allocated into three groups: control group(group A),low-dose silymarin group(group B) and high-dose silymarin group(group C).Four weeks after drug administration,common carotid arteries were removed to determine plaque area,cap thickness and lipid.Serum lipid,malondialdehyde and superoxide dismutase were measured as well. Result:①There were no significant differences in serum lipid between group A and B.Compared with other groups,group C had a lower level of serum lipid(P0.05).②Compared with group A,plaque area and lipid were decreased in group B and C(P0.05) while cap thickness increased(P0.05).Furthermore the changes of group C were more significant(P0.05).③Serum malondialdehyde was lower in group B and C(P0.05) than that in group A,in the meanwhile superoxide dismutase was higher(P0.05).In addition the changes of group C were more significant(P0.05). Conclusion:Silymarin can evidently inhibit the progress of existing atherosclerotic lesions in ApoE-/-mice.The possible mechanisms are lowering serum lipid levels and inhibiting oxidative stress.
