摘  要:目的:利用双向凝胶电泳技术,寻找小鼠心肌梗死后心脏组织蛋白质表达的差异,及大黄素对其靶点蛋白质的影响。方法:建立小鼠心肌梗死模型,腹腔注射大黄素进行干预。利用双向凝胶电泳分离正常小鼠(假手术组)、心肌梗死组及大黄素干预组心脏组织的总蛋白样品。应用凝胶分析软件,分析图谱中差异表达的蛋白质点。结果:各组间蛋白质斑点发生显著性变化。与假手术组相比,心肌梗死组心肌组织中异常表达蛋白质点55个,表达下调的2个(P1,P2),表达上调的53个;大黄素干预组P1蛋白质点明显上调。结论:大黄素影响心肌梗死后心脏组织蛋白质的表达,差异蛋白分离为进一步研究大黄素对心肌梗死后靶点蛋白质的调控机制奠定了基础。Objective:To explore the target protein expressions on mice heart tissues treated by Emodin using two-dimensional gel electrophoresis(2-DE). Method:Myocardial infarction(MI) model in mice was produced and Emodin treated them.The differentially expressed proteins were detected by 2-DE in sham group,MI group and Emodin group. Result:Fifty-five differentially expressed proteins were detected in MI group.Fifty-three proteins of them were up-regulated and two proteins were down-regulated.Whereas one of the down-regulated two proteins was up-regulated in Emodin groups. Conclusion:Emodin modulates protein expression of heart tissue after MI,which may be associated with myocardial injury and protection.
