摘  要:目的:用脂多糖(lipopolysaccharide,LPS)建立阿尔茨海默病(Alzheimer's disease,AD)炎症模型大鼠,观察淫羊藿苷(Icariin ICA)对该模型大鼠脑海马组织内TNF-amRNA、IL-6 mRNA表达的影响并探讨其可能的作用机制。方法:经八臂迷宫训练成功的45只雄性Wistar大鼠随机分为对照组、模型组、ICA低剂量组、ICA高剂量组、双氯芬酸组,采用侧脑室注射LPS制作实验性AD炎症模型,制模后对照组和模型组给予灌胃蒸馏水,双氯芬酸组给予双氯芬酸钠,高、低剂量组给予相应剂量的ICA混悬液后对上述动物进行为期15 d的八臂迷宫测验并记录其数据。迷宫测验完成后断头取脑海马组织,提取RNA,应用SYBR GREENⅠ荧光定量PCR检测实验性AD模型大鼠大脑海马组织内TNF-amRNA、IL-6 mRNA表达。用SPSS统计学软件进行统计学分析。结果:SYBR GREENⅠ荧光定量PCR检测显示:与模型组比较,高剂量组和双氯芬酸组大鼠海马内TNF-amRNA、IL-6 mRNA的表达明显减低(P=0.000)。结论:高剂量ICA显著降低TNF-amRNA、IL-6 mRNA表达。低剂量ICA未产生明显影响,其作用机制可能是通过抑制炎症实现的。Objective: To establish Alzheimer' s disease ( AD ) inflammation model of rats using lipopolysaccharide(LPS). Then to explore the expression and mechanism of TNF-amRNA,IL-6 mRNA in those rats. Methods: After successfully trained in the 8 - arm maze, a total of 45 male Wistar rats were randomly divided into 5 groups, including control group, model group, diclofenac group, low dose group, high dose group. The AD model rats were established by injection LPS into the lateral cerebral ventrical, then the rats in control group and model group were filled the stomach by distilled water, diclofenac group filled diclofenac, the low and high group were filled the corresponding dosage the ICA. Carries on 8-arm maze to the above animal to examine and to record its data for 15 days. Then the brain tissue of rats were removed and withdraws RNA, SYBR GREEN fluorescence quantitative RT- PCR was used to study the expression of TNF-amRNA,IL-6 mRNA. Results:SYBR GREEN I fluorescence quantitative PCR tests showed that diclofenac in rats hippocampus TNF-amRNA, :Compared with model group, high- dose group and IL-6 mRNA expression was significantly reduced. (P= 0. 000 ). Conclusions: high-dose ICA significantly reduced TNF-amRNA, IL-6 mRNA expression, lowdose ICA does not have a significant effects, and its mechanism may be through inhibiting the inflammation.
