摘  要:目的:探讨黄芪联合胰岛素对糖尿病大鼠胰岛素抵抗的作用,并阐明其作用机制。方法:链脲佐菌素(STZ)诱导糖尿病大鼠模型,将诱导成功的糖尿病大鼠随机分为模型组(DM)、黄芪组(RA)、黄芪联合胰岛素组(RA+Ins)和胰岛素组(Ins),同时设立正常大鼠为对照组(Control)。对照组和DM组大鼠无任何处理因素;RA组大鼠给予每日黄芪灌胃;Ins组大鼠每日胰岛素注射;RA+Ins组大鼠给予黄芪灌胃和胰岛素注射。8周后测定大鼠空腹血糖(FBG)值和空腹胰岛素(FINS)水平。腹主动脉取血,检测大鼠血清丙二醛(MDA)含量、超氧化物歧化酶(SOD)活力、肿瘤坏死因子α(TNF-α)含量及大鼠骨骼肌胰岛素受体底物-1(IRS-1)和p38MAPK的表达水平。结果:DM组、RA组、Ins组和RA+Ins组大鼠FBG水平显著高于对照组(P<0.05),RA组、Ins组和RA+Ins组大鼠FBG水平显著低于DM组(P<0.05),Ins组和RA+Ins组大鼠FBG水平显著低于RA组(P<0.05),并且RA+Ins组大鼠FBG水平高于Ins组(P<0.05)。Ins组、RA+Ins组大鼠FINS水平显著高于对照组、DM组和RA组(P<0.01),RA+Ins组大鼠FINS水平显著低于Ins组(P<0.01)。DM组、RA组、Ins组和RA+Ins组大鼠血清MDA含量显著高于对照组(P<0.05),RA组、Ins组和RA+Ins组大鼠血清MDA含量明显低于DM组(P<0.05),RA组和Ins组大鼠血清MDA含量明显高于RA+Ins组(P<0.05)。DM组、RA组、Ins组和RA+Ins组大鼠SOD活力显著低于对照组(P<0.05),RA组、Ins组和RA+Ins组大鼠血清SOD活力显著高于DM组(P<0.05),RA组和Ins组大鼠血清SOD活力显著低于RA+Ins组(P<0.05)。DM组、RA组、Ins组和RA+Ins组大鼠血清TNF-α含量显著高于对照组(P<0.05),RA组、Ins组和RA+Ins组大鼠血清TNF-α含量明显低于DM组(P<0.05),RA+Ins组大鼠血清TNF-α含量明显低于RA组和Ins组(P<0.05)。DM组、RA组、Ins组和RA+Ins组大鼠IRS-1表达低于对照组(P<0.05),RA+Ins组大鼠IRS-1表达高于DM组、RA组和Ins组(P<0.05)。DM组、RA组、Ins组和RA+Ins组大鼠p38MAPK�Objective To explore the effect of astragalus combined with insulin on insulin resistance in diabetic rats and to clarify the action mechanism.Methods The diabetic rat models induced by STZ were randomly divided into diabetic model(DM) group,astragalus(RA) group,insulin(Ins) group and astragalus combined with insulin(RA+Ins) group.At the same time the normal rats were used as control group.After treatment for 8 weeks,the levels of fasting blood glucose(FBG) and fasting insulin(FINS) of rats were measured,and the contents of MDA and TNF-α in serum were detected,and the levels of insulin receptor substrate-1(IRS-1) and p38MAPK in skeletal muscle were examined by Western blotting.Results Compared with control group,the levels of FBG of rats in DM,RA,Ins and RA+Ins groups were significantly increased(P0.05),and the FBG levels in RA,Ins and RA+Ins groups were significantly decreased compared with DM group(P0.05);the FBG level in RA+Ins group was significantly decreased compared with RA and Ins groups(P0.05).Compared with DM,RA and control groups,the levels of FINS in Ins and RA+Ins groups were increased(P0.05),and the FINS level in RA+Ins group was decreased compared with Ins group(P0.05).Compared with control group,the levels of MDA and TNF-α in DM,RA,Ins and RA+Ins groups were significantly increased(P0.05),and they were lower in RA,Ins and RA+Ins groups compared with DM group(P0.05);compared with RA and Ins groups,the MDA and TNF-α in RA+Ins were decreased(P0.05).The activities of SOD in DM,RA,Ins and RA+Ins groups were significantly decreased compared with control group(P0.05),and they were higher in RA,Ins and RA+Ins groups compared with DM group(P0.05);compared with Ins and RA groups,it was increased in RA+Ins group(P0.05).The expression levels of IRS-1 in DM,RA and RA+Ins groups were decreased compared with control group(P0.05),but it was higher in RA+Ins than DM,RA and Ins groups(P0.05).The expression
