摘 要:目的探讨哮喘大鼠肺组织中细胞周期蛋白D1(Cyclin D1)的表达对气道重塑的影响。方法将20只大鼠随机分为正常组(10只)和哮喘组(10只)。哮喘组分别于1 d和8 d用卵蛋白(OVA)10 mg、氢氧化铝20 mg和生理盐水2 ml制成悬液腹腔注射致敏。15 d开始用1%OVA雾化吸入激发,每周3次,共8周。末次激发24 h后处死,取右下肺组织用10%甲醛固定、石蜡包埋和切片。用图像分析仪测定气道内周径(Pi)、外周径(Pe),并计算管壁面积(WA)、气道平滑肌面积(SMC-A)、气道平滑肌细胞数(N),且WA、SMC-A、N均用Pi进行标准化。免疫组化检测肺组织中CyclinD1的表达。正常组大鼠用生理盐水替代卵蛋白,余同哮喘组。结果哮喘组肺组织中Cyclin D1的表达、WA/Pi、SMC-A/Pi和N/Pi均高于正常组,差异均有统计学意义。CyclinD1与WA/Pi、SMC-A/Pi和N/Pi均呈正相关性。结论哮喘大鼠肺组织中cyclin D1高表达参与气道平滑肌细胞增殖,从而导致哮喘气道重塑。Objective To investigate Cyclin D1 expression in pulmonary tissue and its effect on airway remod- eling in asthmatic rats. Methods Twenty rats were randomly divided into normal groups and asthma groups with rats ten in each group. Asthma groups rats were injected ovalbumin (OVA) 10 mg each, made of aluminum hy- droxide 20 mg suspension in place of 2 ml Saline by intraperitoneal respectively in 1day and 8day. Asthma groups rats were excitaed by aerosoling inhalation with 1% OVA, and there were three times a week for 8 weeks. Rats were killed after Last excitation 24 h and right lower part of pulmonary tissue was fixed with 10% formalin, embe- ded with paraffin and sected. Using image analysis to test airway circumference (Pi), outer circumference (Pe), and calculate the wall area (WA), airway smooth muscle area (SMC - A), airway smooth muscle cells (N), and WA, SMC - A, N were standardized by Pi. Expression of Cyclin D1 in pulmonary tissue were detected by lm- munohistochemical. Ovalbumin were raplaced by salinein in normal groups rats, and other contents were as same as asthma groups. Results Expression of Cyclin D1 in pulmonary tissue ,WA/Pi, SMC - A/Pi and N/Pi in the asthma group was significantly higher than the normal group. Cyclin D1 and WA/Pi or SMC - A/Pi or N/Pi showed a positive correlation. Conclusion High expression of cyclin D1 participated airway smooth muscle cell proliferation, resulte in airway remodeling in asthma rats.
