黄芪失笑散对急性心肌梗死大鼠心功能及Caspase-8表达的影响

摘　　要：目的观察不同剂量黄芪失笑散对大鼠急性心肌梗死后心功能及凋亡相关蛋白Caspase-8表达的影响。方法 120只雄性SD大鼠随机分为5组:模型组、黄芪失笑散大剂量组、中剂量组、小剂量组、对照组。连续灌胃7 d后,结扎冠状动脉左前降支造成急性心肌梗死模型,对照组不结扎,余步骤相同,手术后4.5 h检测相关指标。颈动脉插管检测血流动力学指标[左室内压峰值(LVSP)、左室等容收缩末期压力上升最大速度(+dp/dtmax),左室舒张末压力(LVEDP)、左室压力下降最大速度(-dp/dtmax)],Western blot法检测心肌梗死边缘区Caspase-8蛋白表达量。结果与模型组比较,黄芪失笑散组可明显改善心肌梗死大鼠血流动力学指标,下调Caspase-8蛋白表达量,且呈剂量依赖性,以大剂量组效果最好,各剂量组之间差异无统计学意义。结论黄芪失笑散能够改善心梗后大鼠的心功能,并降低梗死边缘区凋亡相关蛋白Caspase-8水平。Objective： To investigate the effects of different dosages of Huangqishixiao Powder on cardiac function and protein expression levels of Caspase-8 in rats with acute myocardial infarction.Methods： 120 male SD rats were randomly divided into 5 groups： the model group,the large-dose of Huangqishixiao Powder group,the medium-dose of Huangqishixiao Powder group,the low-dose of Huangqishixiao Powder group and the sham group.After being administered correspnoding medicine for 7 days,all groups excepts sham group underwent coronary ligation of left anterior descending branch to establish acute myocardial infarction models.However,the sham group only received thoracotomy without coronary ligation.Four and half hours after AMI,cardiac function（LVSP,LVEDP,＋dp/dtmax,-dp/dtmax） and protein expression levels of Caspase-8 were measured by western blot method.Results： Comparing with rats in model group,the LVSP,＋dp /dtmax and-dp/dtmax were higher in the Huangqishixiao Powder group.However,the LVEDP and the expression levels of Caspase-8 were lower in the Huangqishixiao Powder group.The effect of the large-dose of Huangqishixiao Powde group was the best in these groups.Conclusion： Huangqishixiao Powder could improve cardiac function and decrease the expression levels of Caspase-8 relected with apoptosis.
