摘 要:目的研究中药复方清热颗粒对耐环丙沙星鲍氏不动杆菌外排泵耐药机制的影响。方法建立急性耐环丙沙星鲍氏不动杆菌腹腔感染兔动物模型,共8只,并设对照组大耳白兔6只。造模成功后,模型组予复方清热颗粒药液灌胃,空白对照组则给予同等体积的蒸馏水灌胃,连续给药5 d。末次给药后无菌心脏取血,离心分离出含药血清和空白血清,-20℃冰箱保存备用。应用外排泵抑制剂(CCCP)筛选出外排泵阳性的耐环丙沙星鲍氏不动杆菌8株,利用琼脂对倍稀释法测出上述8株鲍氏不动杆菌对环丙沙星的最小抑菌浓度,用同样的方法测出在分别联合含药血清、空白血清、复方清热颗粒、CCCP的情况下环丙沙星对上述8株鲍氏不动杆菌的最小抑菌浓度(MIC)变化。结果中药复方清热颗粒含药血清可以降低环丙沙星对鲍氏不动杆菌的MIC值。结论复方清热颗粒含药血清可抑制鲍氏不动杆菌的外排泵,从而改善鲍氏不动杆菌对环丙沙星的耐药。Objective: To explore Compound Qingre Granules on efflux pumps of acinetobacter baumannii which was resistant to Ciprofloxacin.Methods: The model group including 8 rabbits were to be intraperitoneal injection CIP-resistant Acinetobacter baumannii to create the acute abonomal infection rabbit models.And the control group had 6 rabbits.The model group was taken Compound Qingre Granules by intragastric administration for 5 days.The control group was taken water in the same way and the same dose for 5 days.After the last drug,the drug serum and the blank serum were separated from heart blood by centrifugation and saved in refrigerator-20℃.Application of efflux pump inhibitor(CCCP) was used to choose 8 strains positive efflux pump CIP-resisitant acinetobacter baumannii.The minimal inhibitory concentration of Ciprofloxacin was measured when it was combining with drug serum,blank serum,Compound Qingre Granules and CCCP to the 8 strains acinetobacter baumannii by agar dilution method.Results: Compound Qingre Granules containing drug serum reduced the MIC of ciprofloxacin to acinetobacter baumannii.Conclusion: The serum of Compound Qingre Granules can inhibit efflux pump of the CIP-resistant Acinetobacter baumannii and improve the resisitance of Acinetobacter baumannii to CIP.
