慢性肾功能衰竭肾阳虚型大鼠肾组织细胞糖皮质激素受体、细胞白介素-1β及肿瘤坏死因子-α的变化

摘　　要：目的研究肾阳虚型慢性肾功能衰竭(CRF)大鼠肾组织细胞糖皮质激素受体(GR)及白介素-1(IL-1β)、肿瘤坏死因子-α(TNF-α)等炎性因子的变化,探讨CRF肾阳虚证的证候学基础。方法采用腺嘌呤灌胃复制肾阳虚型CRF模型,并设正常组、模型组(腺嘌呤每日200 mg/kg)、济生肾气丸组(腺嘌呤每日200 mg/kg+济生肾气丸每日40 g/kg)、保肾康组(腺嘌呤每日200 mg/kg+保肾康每日33.3 mg/kg),保肾康为对照组。实验全过程24 d,第1～12日每日灌胃1次,第12日后隔日灌胃1次。实验结束,用酶联免疫吸附法测定肾组织IL-1β、INF-α及GR的水平。结果模型组大鼠肾组织细胞IL-1β、INF-α水平明显升高,GR水平明显降低。济生肾气丸组、保肾康组与模型组比较IL-1β、INF-α水平显著降低,GR水平显著升高。济生肾气丸组同保肾康组比较IL-1β、INF-α明显降低,GR水平明显升高。结论 IL-1β、INF-α及GR与CRF肾阳虚证密切相关;济生肾气丸能降低IL-1β、TNF-a水平,并能提高GR的活性。Objective： To investigate the effect of glucocorticoid receptor（GR）,Interleukin-1β（IL-1β） and tumor necrosis factor alpha（TNF-α） in chronic renal failure（CRF） with kidney-yang deficiency.Methods： CRF model with kidney-yang deficiency was replicated by adenine infused into stomach.SD rats were randomly divided into the normal group,the model group with adenine 200 mg/kg one day,the Jishengshenqi Decoction group with adenine 200 mg/kg plus Jishengshenqi Decoction in 40 g/kg one day,and the Baoshenkang group with adenine 200 mg/kg plus Baoshenkang 33.3 mg/kg a day.The whole process was 24 days.From the 1st day to 12th day,the gastric perfusion was once a day.After the 12th day,the gastric perfusion was alternate days once.The contents including IL-1β,INF-α and GR were determined by ELISA.Results： Compared with the normal group,the contents of IL-1β、INF-α of kidney tissues were increased significantly in the model group.The content of GR in kidney tissues was decreased significantly in the model group.Compared with model group,the contents such as IL-1β,TNF-α of kidney tissues were decreased remarkably in Jishengshenqi Decoction group and Baoshenkang group.The contents of GR were increased obviously in Jishengshenqi Decoction group and Baoshenkang group.Compared with Baoshenkang group,the contents of IL-1βand TNF-α were decreased significantly in Jishengshenqi Decoction group.The content of GR were increased significantly in Jishengshenqi Decoction group.Conclusion： IL-1β、TNF-α and GR were related to CRF with kidney-Yang deficiency closely.Jishengshenqi Decoction decreased the content of IL-1βand TNF-α of kidney tissues and increased the content of GR.
