摘  要:目的观察杞藿饮对力竭游泳大鼠肝细胞衰老及其超微结构的影响。方法 48只雄性SD大鼠按数字表法随机分为4组,分别为游泳对照组和杞藿饮低、中、高剂量组,每组12只。3周后检测各组大鼠血清一氧化氮(NO)、丙氨酸氨基转移酶(ALT)和天门冬氨酸氨基转移酶(AST)水平,比较各组肝细胞H2A磷酸化染色及超微结构变化。结果高、中、低浓度杞藿饮组血清NO、ALT、AST水平与游泳对照组比较均明显降低(P<0.05),高浓度和中浓度杞藿饮组血清NO、ALT、AST水平与低浓度组比较降低更明显(P<0.05)。3个杞藿饮组肝细胞H2A磷酸化阳性细胞数均明显少于游泳对照组(P<0.05),肝细胞超微结构表明损伤程度明显轻于游泳对照组,且与浓度增加呈正相关。结论杞藿饮能明显缓解大鼠力竭游泳造成的肝脏损害,可能与降低肝细胞衰老进程有关。Objective To investigate the effects of Qihuo tonic (QHT) on hepatic cell aging and uhrastructure in swimmingexhausted rats. Methods Sixty male SD rats were randomly divided into 4 groups : the swimming-exhausted group, the low dose QHT group, the moderate dose QHT group and the high dose QHT group. After 3 weeks, changes in serum nitrogen monoxidum ( NO), ala- nine aminotransferase (ALT) and aspartate amino transferase (AST) of all the groups were determined. Then, HA2 phosphorylation staining was used to observe hepatic cell aging, and the uhastructural changes in hepatic cells were compared between the groups. Results Compared with those of the swimming-exhausted group, the levels of NO, ALT and AST in the high, moderate and moderate dose QHT groups decreased significantly ( P 〈 0. 05 ) , the levels of NO, ALT and AST of the high and low dose groups decreased more significantly, when compared with those of the low dose group (P 〈 0. 05 ). The number of H2A staining positive hepatic cells of the 3 QHT groups was significant lower than that of the control group (P 〈 0. 05). The extent of damage to the ultrastructure of hepatic cells in the 3 QHT groups was obviously lighter than that of the control group, and positive correlation could be noted between damage extent and QHT concentration. Conclusion QHT could obviously alleviate hepatic damage induced by exhausted fatigue in rats, the mechanism of which might be related with the decreased process of hepatic cell aging.
