姜黄素对于大鼠脑出血后ICAM-1表达的影响

摘　　要：目的探讨脑出血(ICH)后血肿周围组织细胞黏附分子(ICAM-1)含量的变化及早期姜黄素治疗对其表达的影响。方法采用未抗凝新鲜自体股动脉血注入大鼠尾状核建立脑出血动物模型。将60只大鼠随机分成对照组、ICH组、姜黄素低干预组,于建模后给予300mg/(kg.d)姜黄素,连续腹腔注射7天,各组分6小时、1天、3天和7天共4个时间点。分别采用干重湿重法、免疫组织化学法测定不同时间点的脑组织水含量和ICAM-1表达的变化。结果脑组织水含量和ICAM-1表达均在ICH后6小时开始升高,3天达到高峰,7天下降至正常水平,两者之间存在明显的相关性(r=0.88,P<0.01)。姜黄素小剂量及中等剂量治疗能够降低各个时间点的脑组织水含量和ICAM-1的表达。结论 ICAM-1与脑水肿的形成密切相关;姜黄素治疗可以降低ICH后血肿周围脑组织ICAM-1的表达,减轻ICH后的脑水肿形成。Objective To investigate dynamic changes of water content and ICAM-1in Intracerebral hemorrhagic（ICH） rats and the role that ICAM-1plays in cerebral edema after hemorrhage,and to evaluate the effect of curcumin on reducing the content of cerebral edema and ICAM-1expression.Methods Animal model was made by injecting self arterial blood into the caudate nucleus of rats.60rats were randomly assigned to control group,cerebral hemorrhage group and curcumin intraperitoneal injection treated group.After the injection,the rats were sacrificed at 6h,1d,3d,7drespectively.Brain water content was determined by wet-dry weight,while the expression of ICAM-1was detected by immunohistochemical method over a time course ranging from 6hto 7d.Results Brain water content and ICAM-1increased after 6hof cerebral hemorrhage and reached its climax at the 3d,then decreased to the normal level at the 7d.And there is an obvious correlation between them（r=0.88,P〈0.01）.Administration of curcumin could reduce the content of brain edema and ICAM-1expression.Conclusion Curcumin can reduce ICAM-1expression,thus can reduce cerebral edema after ICH.These suggest a close correlation between ICAM-1 and cerebral edema,and curcumin might be available for the therapy of cerebral edema after ICH.
