摘 要:目的探讨阿仑膦酸钠对骨量和骨质量的作用。方法将48只12月龄SD大鼠随机分为去卵巢模型组(OVX)、假手术组(SHAM)、低剂量阿仑膦酸钠组(ALL)、高剂量阿仑膦酸钠组(ALH),每组12只。ALL与ALH组大鼠作双侧卵巢切除,分别给予阿伦膦酸钠0.5mg/(kg·d)与1.0mg/kg(kg·d),持续用药12个月。结果 ALL及ALH组大鼠尿钙与肌酐比值(Ca/Cr)均低于OVX组,差异有统计学意义(P<0.05);ALL及ALH组尿羟脯氨酸与肌酐比值(HOP/Cr)与血清碱性磷酸酶水平(ALP)均较OVX组低,差异有统计学意义(P<0.05);SHAM组骨干重、灰重和钙含量高于OVX组,ALL与ALH组高于OVX组,差异均有统计学意义(均P<0.05);ALL与ALH组股骨骨密度、抗弯力及全身骨密度均高于OVX组(P<0.05)。结论 SD大鼠卵巢切除12个月后出现明显骨质疏松,阿仑膦酸钠干预对增加骨量、改善骨质量有显著作用,在一定范围内骨质量的改善呈剂量依赖性。Objective To investigate the effect of Alendronate on bone mass and bone quality in oophorecto- mized rats. Methods 48 SD rats aged 12 months were randomly divided into OVX (ovariectomized), SHAM (sham- operated) group, ALL (Alendronate low dosage) group, and ALH (AIendronate high dosage) group. Bilateral oopho- rectomy was performed in the ALL and ALH groups. At the same time, the two groups expriment rats were given Alendr- onate in the dosage of 0. 5 mg/kg/day and 1.0 mg/kg/day for 12 months. Results The urinary calcium/creatinine ratio was significanly lower in Alendronate-treated OP model groups than that in OVX group (P 〈 0. 05 ). The urinary hydroxyproline/creatinine ratio and serum alkaline phosphatase level showed a decreasing tendency in Alendronate-treated OP model groups as compared with OVX group (P 〈 0. 05). Dry bone weight, bone ash weight, bone calcium content in SHAM group were higher than OVX group, and those in the ALL and ALH groups were higher than OVX group (P 〈 0. 05). The femur bone mineral density, anti-bend strength, and the body bone mineral density in ALL and ALH was increased in alendronate than that in OVX group (P 〈 0. 05). Conclusion The oophorectomized SD rats showed clearly osteoporosis 12 months groups after operation. Alendronate can increase bone mass, improve bone quality, which is dose-dependent in a certain range of bone quality improvement.
