摘 要:目的制备石蒜碱脂质纳米粒,考察体外抗肿瘤活性。方法制备石蒜碱-油酸复合物,溶剂注入法制备纳米乳,超滤法测定包封率,MTT法初步考察纳米乳的抗肿瘤活性。结果制备的纳米乳颗粒圆整均匀,形态良好,包封率为82%±1.4%,粒径为91.9±4.0 nm,PDI为0.171±0.013,Zeta电位为-50.4±5.4 mV。纳米乳较原药能更好抑制肿瘤细胞增殖。结论溶剂注入法制备的石蒜碱脂质纳米乳包封率较高,粒径大小分布均匀,较原药具有更强的抗肿瘤活性。OBJECTIVE This study was to prepare a lycorine lipid nano - emulsion formulation for the increased antitumor activity in vitro. METHODS Lycorine - oleic acid loaded lipid nano - emulsions were prepared by solvent injection method and characterized by transmission electron microscopy (TEM), particle size,polydispersity index, Zeta potential and entrapment efficiency analysis. The in vitro growth inhibition activity studies were performed on A549 cell lines by MTT test. RESULTS The droplets were spherical and uniform. The entrapment efficiency was 82% ±1.4qv , size was 91.9±4.0 nm, PDI was 0.171 ± 0.013, and Zeta potential was - 50.4 ± 5.4 inV. The effective test by MTT assay showed better growth inhibition activity of lycorine nanoemulsion on A549 cell lines when compared with lycorine. CONCLUSION Lycorine nanoemulsion showed high entrapment efficiency and a narrow size distribu- tion. Lycorine nanoemulsion showed better antitumor activity than free drug.
