摘 要:目的明确坤宁安治疗围绝经期综合征的分子机制。方法将13~16月龄雌性Wist-ar大鼠40只随机分为四组,分别为模型组、坤宁安高剂量组、坤宁安低剂量组、西药对照组,另选3~5月龄雌性Wistar大鼠为青年对照组,每组10只。应用RT-PCR方法检测各组大鼠卵巢丝裂原活化蛋白激酶(MAPK)、增殖细胞核抗原(PCNA)、细胞外调节蛋白激酶(ERK1/2)mRNA表达水平。结果青年对照组、坤宁安高剂量组、坤宁安低剂量组大鼠卵巢MAPK、PCNA、ERK1/2 mRNA表达升高,与模型组比较,差异有统计学意义(P<0.05)。西药对照组与模型组相比MAPK、PCNA mRNA表达水平比较,差异无统计学意义(P>0.05);ERK1/2 mRNA表达降低,差异有统计学意义(P<0.05)。结论坤宁安治疗围绝经期综合征的作用机制与有效调节卵巢分裂增殖通路、促进卵巢细胞增殖相关。Objective To determine the molecular mechanism of kunningan in the treatment of peri- menopausal syndrome. Methods Forty female Wistar rats, aged 13 - 16 months were randomized into four groups, named a model group, a high - dose kunningan group, a low - dose kunningan group and a western medicine control group. Additionally, a youth control group of female Wistar rats aged 3 - 5 months was set up. Ten rats were in each group. RT - PCR method was adopted to detect the mRNA expressions of ovarian mitogen activated protein kinase(MARK) , proliferating cell nuclear antigen(PCNA) and extracellular regula- ting protein kinase ( ERK1/2 ) in the rats. Results In the youth control group, the high - dose kunningan group and the low- dose kunningan group,the mRNA expressions of MARK,PCNA and ERK1/2 in the rat ovaries were increased, presenting the statistically significant differences as compared with the model group (P 〈0.05). The differences in the mRNA expressions of MARK and PCNA were not significant statistically(P 〉 0.05 ) , but the difference in ERK1/2 mRNA expression was significant statistically ( P 〈 0.05 ) between the western medicine control group and the model group. Conclusion The mechanism of kunningan is related to the effective regulation of ovarian division and proliferation pathways and the promotion of ovarian cellular proliferation in the treatment of perimenopausal syndrome.
