摘 要:目的:观察蒙药五味风湿胶囊对佐剂性关节炎的作用及其免疫调节机制。方法:取Wistar大鼠,分成正常组、佐剂性关节炎(AA)模型组、五味风湿胶囊低剂量组(0.2 g.kg-1.d-1)、中剂量组(0.4 g.kg-1.d-1)、高剂量组(0.8 g.kg-1.d-1)及忠伦-5汤组(原剂型1.68 g.kg-1.d-1),测定各组大鼠关节肿胀程度,关节组织白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、前列腺素(PGE2)及一氧化氮(NO)水平,测定血清超氧化物歧化酶(SOD)活性和丙二醛(MDA)水平。通过细胞培养,考察药物对AA大鼠脾细胞增殖能力的影响,探讨该药干预下AA大鼠腹腔巨噬细胞培养液对C57BL/6J小鼠胸腺细胞增殖能力的影响。结果:五味风湿胶囊可抑制AA大鼠关节组织IL-1β,TNF-α,PGE2,NO水平;提高血清SOD活性,但MDA未见显著变化;可抑制AA大鼠脾细胞增殖能力;抑制AA大鼠腹腔巨噬细胞培养液对C57BL/6J小鼠胸腺细胞增殖能力的影响。结论:五味风湿胶囊具有良好的抗佐剂性关节炎作用。其作用机制可能与抑制相关细胞因子,调节相关酶的活性及免疫器官细胞增殖等有关。Objective: To study the effects and immunoregulation mechanism of the traditional Mongolian medicine Wuweifeng- shi capsule on adjuvant arthritis (AA). Method: Wister rats were divided into several groups: normal group, AA model group, Wu- weifengshi capsule groups ( with low, moderate, high dose of 0. 2, 0. 4,0. 8 g·kg^-1·d^-1 respectively) , and Zhonglun-5 group (original dose of 1.68 g ·kg^-1·d^-1 ). The edema degree, the level of IL-1β, TNF-α, PGE2, NO and MDA and the activity of SOD in serum were detected. Through cell cuhure, the effects of the medicine on AA rat's splenic cell's multiplication capacity were stud- ied. The influence of celiac macrophage cell culture fluid of AA rats' on C57BL/6J mice thymic cell multiplication capacity under the medicine was evaluated. Result: Wuweifengshi capsule showed an inhibiting function on the level of IL-1β, TNF-α, PGE2 , NO and in- creased the activity of SOD in serum, but showed no significant influence on MDA. It also inhibited the AA rat's splenic cell's multi- plication capacity and the influence of celiac macrophage cell culture fluid of AA rat's on C57BL/6J mice thymic cell multiplication ca- pacity. Conclusion : The anti-AA effect of Wuweifengshi capsule is possibly due to its inhibition of relevant cytokines and its adjust- ment of corresponding enzyme's activity and immunization organ's cell multiplication capacity.
