摘 要:分泌蛋白是一类重要的介导病原与宿主相互作用的因子。微孢子虫专性细胞内寄生的生活方式决定了分泌蛋白在其侵染、增殖、防御和调控宿主生理活动中具有重要的作用。利用真核生物分泌蛋白预测流程EuSecPred 1.0对家蚕微孢子虫全基因组的分泌蛋白基因进行预测和严格条件筛选,鉴定获得97个分泌蛋白基因,占基因组全部编码基因的1.87%。对分泌蛋白的功能和结构域进行注释分析,发现了孢壁蛋白、丝氨酸蛋白酶抑制剂(Serpin)和Ricin B-凝集素等重要侵染相关因子。氨基酸组成分析表明,家蚕微孢子虫分泌蛋白及其信号肽主要由疏水性氨基酸组成,而信号肽剪切位点主要由亲水性氨基酸组成。基序分析发现,分泌蛋白信号肽中存在保守基序T[ILV][IVL][LI][VIL][LI]LS[IV]I[AK][SA],分泌蛋白内部存在[ES]W[VLM]K和YF[FY][DG]L 2种基序。在全基因组范围对家蚕微孢子虫的分泌蛋白进行预测分析,有助于进一步研究家蚕微孢子虫致病相关的候选因子。Secreted proteins are important factors mediating the interaction between pathogen and host.The obligate intracellular parasitism of microsporidia indicates an important role of secreted proteins during the infection,proliferation,defense and regulating physiological activity of the host.Here,at whole genome level,we conducted prediction and stringent screening for secreted proteins of the Nosema bombycis with EuSecPred 1.0 pipeline and identified 97 secreted proteins,accounting for 1.87% of its all encoded proteins.Some important infection related proteins including spore protein,serine protease inhibitor(Serpin) and ricin B-lectin were identified through annotation and analysis to functional and structural domains of the secreted proteins.Amino acid composition analysis indicated that the secreted proteins and the signal peptides of Nosema bombycis are mainly composed of hydrophobic amino acids,but the splice sites of signal peptides are mainly composed of hydrophilic amino acids.Motif analysis revealed a conserved motif,TT[ILV][IVL][LI][VIL][LI]LS[IV]I[AK][SA],in signal peptides and two conserved motifs,WK and YF[FY][DG]L,inside the sequences of secreted proteins.By this study we firstly predicted and analyzed the secreted proteins of N.bombycis at whole genome level,providing good references for studying candidate factors related to pathogenicity of N.bombycis.
