摘  要:目的:探讨腺嘌呤按100 mg/100 g给药10 d后大鼠生殖毒性及肝肾毒性的时效性。方法:将上述25只动物购入后,正常饲养1周,按体重均匀分组,共5组。每组5只,第1组为空白组,2 mL/(kg.d)剂量蒸馏水灌胃;第2~5组为给药组,将腺嘌呤按50 mg/mL浓度配制,按与腺嘌呤1∶10比例加入阿拉伯胶作为助溶剂,2 mL/100(g.d)剂量灌胃10 d,分别在给药0、17、30、46 d各处死1组,分析精子质量、称量体重及器官重量、用放免法检测性激素及肝肾功能的变化,并对腺嘌呤生殖毒性的时效和量效关系进行比较分析。结果:在给药10d结束后,腺嘌呤所产生的生殖毒性和肝肾毒性仍在延续,死亡率随着饲养时间的延长而增加,当46 d只有1只大鼠存活。结论:腺嘌呤法制作雄性Wistar大鼠肾阳虚型不育症动物模型最佳剂量点应<100 mg/100(g.d)和时间点<10 d。Objective:To study the chronergy of the rat~ reproductive toxicity and liver and kidney toxicity after admin-istration of adenine with a dose of 100 mg/100 g for 10 days. Methods :25 animals were normally fed for one week. Ac- cording to the weight ,they were evenly divided into five groups, 5 in each group. The first group was the control group, the dose gavage was 1 mL/( kg · d) distilled water; group 2 to group 5 were the administration groups, the preparation of the adenine was 50 mg/mL, adding ratio of 1 : 10 with adenine gum arabic as cosolvent, 1 mL/100 ( g · d) for gavage 10 days, rats were killed at the 0th 17th 30th and 46th day after administration respectively and analysis of sperm quality, weights and organ weights, sexhormones was carried on by radioimmunoassay and changes in the fiver and kidney func- tion, and the aging of adenine reproductive toxicity and dose - effect relationship were observed for comparative analysis. Result:lO days after administered, the adenine reproductive toxicity and liver and kidney toxicity still continued, the mor- tality rate was increased with the prolonged feeding time, on the 46th day there was only one rat survival. Conclusion : Ade- nine for establishing Wistar rats model of infertility due to kidney Yang deficiency should be used the optimal dose 〈 100 mg/100 (g ·d) and time 〈10 days.
