摘 要:目的:通过桔梗总皂苷对肺炎支原体(MP)的MIC、MBC及TIME-KILL曲线测定,观察其在体外对MP生长抑制及杀灭作用。方法:采用微量倍比稀释法测定MIC和MBC,利用定量PCR检测不同孵育时间,不同浓度药物对MP繁殖速度的影响。结果:桔梗总皂苷体外抗MP的MIC为16~64μg.mL-1;MBC为64~128μg.mL-1;桔梗总皂苷浓度为32到64μg.mL-1时,可以降低MP生长速度并呈现一定的量效关系,当浓度达到128μg.mL-1时可完全抑制MP在培养液中的生长繁殖。结果:桔梗总皂苷在体外具抑制MP生长繁殖作用。Objective : To observe the activity of Platycodon saponins against Mycoplasma Pneumoniea through experi- ments of MIC, MBC and TIME - KILL. Method:MICs and MBCs of platycodon saponins were determined by broth microdi- lution method. MP was incubated in PPLO broth at concentrations equivalent to one, two and four times of the MIC that had previously been determined. The organisms of M. Pneumoniae was incubated at 37℃. The concentrations of MP be- fore and after diferent incubate time were detected by quantitative PCR. Result:MICs of platycodon saponins were 16 -64 μg · mL^- 1 and MBCs were 64 - 128 μg · mL^- 1 against MP in vitro. Replication of MP in PPLO was inhibited by platyc- odon saponins when the concentration was between 32 μg · mL^- 1 and 64μg · mL^- 1. The number of MP in PPLO broth was reduced and the growth was completely inhibited when the concentration of platycodon spaonins reached 128 μg · mL^- 1. Conclusion :Platycodon saponins has anti -M. Pneumoniea activity in vitro.
