摘 要:目的:研究健脾理气方对人胃癌移植瘤小鼠的5-Fu代谢过程的影响。方法:将80只小鼠按照随机分为人胃癌移植瘤模型组、5-Fu化疗组、健脾理气中药组、5-Fu化疗+中药组,每组20只。将MKN-8荷瘤小鼠新鲜癌块磨碎在小鼠腋窝下接种,制造人胃癌移植瘤小鼠模型。通过高效液相色谱法(HPLC法)检测各组小鼠移植瘤组织及血浆中5-Fu的浓度。结果:中药+化疗组小鼠移植瘤组织中的5-Fu浓度高于单纯化疗组(P<0.05);中药+化疗组与单纯化疗组血浆中的5-Fu浓度则无显著差异(P>0.05)。结论:中药健脾理气方能够提高化疗药物5-Fu在荷瘤小鼠肿瘤组织中的浓度,起到增效的作用。Objective : To research the impact of Spleen - fortifying and Qi - regulating formula on the metabolic proces- ses of 5 -Fu in the mice with transplanted gastric cancer cells of human. Methods :80 mice were divided into model group in which the gastric cancer cell of human was transplated ,5 - Fu chemotherapy group, Spleen - fortifying and Qi - regula- ting Formula group and the group using the Spleen - fortifying and Qi - regulating Formula and 5 - Fu chemotherapy, each group contained 20 mice. The animal model was made by grinding the tumor of MKN - 8 tumor - hearing mice and trans- planting the cells of tumor to the axillary of mice. The method of high performance liquid chromatography (HPLC) was used to detect the density of 5 - Fu in tumor and plasma in mice. Results : The density of 5 - Fu of the group using the Spleen - fortifying and Qi - regulating Formula and 5 - Fu chemotherapy in the tumor of mice was obviously higher than that in the chemotherapy group( P 〈0.05 ). There was no difference between Chinese herbal drugs combined with chemotherapy group and the chemotherapy group in the density of 5 - Fu in plasma of mice ( P 〉 0. 05 ). Conclusion : The Spleen - fortifying and Qi - regulating Formula can raise therapeutic effect on the the tumor of mice by raising the density of 5 - Fu in it.
