摘 要:目的:研究当归补血汤加味方对血管性痴呆(vascul ar dement i a,VD)大鼠的脑保护作用及其机制。方法:采用线栓法制成大鼠VD模型。用Morri s水迷宫来检测VD大鼠学习记忆能力的改变,应用免疫组织化学染色法检测VD大鼠海马区神经生长因子(nerve growt h fact or,NGF)的表达,观察当归补血汤加味方对其影响。结果:Morri s水迷宫检测表明VD术后大鼠游泳距离和潜伏期明显延长,而VD术后当归补血汤加味方灌胃组大鼠学习记忆能力有所改善;VD大鼠海马NGF免疫反应阳性细胞分布稀疏,数目比假手术对照组及VD术后当归补血汤加味方灌胃组明显减少(P<0.05);而VD术后大鼠当归补血汤加味方灌胃组与假手术对照组海马区NGF阳性细胞的表达无明显性差异(P>0.05)。结论:VD术后大鼠学习记忆能力下降,而当归补血汤加味方能改善VD大鼠学习记忆能力;VD术后大鼠海马区NGF免疫反应阳性细胞的表达降低,当归补血汤加味方能增强脑VD大鼠海马区NGF阳性细胞的表达,对血管性痴呆大鼠的脑组织具有一定的保护作用。Objective: To study cerebral protection and its mechanism of modified DangGui BuXueTang in treating rats suffering from vascular dementia (VD). Method: VD models were established with suture occluded method. Changes of learning and memory ability of VD rats were detected with Morris water maze and expressions of nerve growth factor (NGF) in the hippocampus of VD rats were measured with immunohistochemical staining method to observe the effects of modified DangGui BuXueTang on the rats. Result: The detection of Morris water maze indicated that swimming distance and incubation period of rats after operation were prolonged, learning and memory ability of rats in intragastric administration group of modified DangGui BuXue Tang after surgery were im- proved; NGF immunoreactive cells in the hippocampus of VD rats were scattering, the number was less than that of sham-operation group and intragastric administration group of modified DangGui BuXue Tang after surgery remark- ably (P〈0.05); while comparison between sham-operation group and intragastric administration group of modified DangGui BuXue Tang after surgery showed no statistical difference in the expressions of NGF positive cells in the hippocampus (P〉0.05). Conclusion: Modified DangGui BuXue Tang could improve the learning and memory ability of VD rats when the ability is lowered after surgery; it could also enhance the expressions of NGF positive cells in the hippocampous and protect cerebral tissue of VD rats when the expression is decreased.
