摘 要:目的建立鉴别荷叶炮制品总生物碱化学成分的高效液相色谱-电喷雾离子阱质谱(HPLC-ESI/MSn)方法,分析炮制对荷叶中生物碱类成分的影响。方法采用HPLC-ESI/MSn技术,分析荷叶生、炭品总生物碱,根据正离子模式下获得的各色谱峰质谱数据,参照文献和荷叶碱对照品对比,对主要色谱峰进行指认。结果鉴别了生荷叶总生物碱中10种生物碱和荷叶炭总生物碱中6种生物碱。荷叶制炭后荷叶碱、N-降荷叶碱、番荔枝碱、甲基莲心碱消失,另外有6种生物碱色谱峰面积显著降低。结论荷叶制炭后生物碱类成分发生明显变化,产生3种未知成分需进一步研究。AIM To establish the HPLC-ESI /MS n method to analyze the changes in total alkaloids in processed product of Nelumbinis Folium.METHODS The total alkaloids in fresh and carbonized Nelumbinis Folium were identified by HPLC-ESI /MS n in positive modes.RESULTS Ten kinds of alkaloids were identified from fresh Nelumbinis Folium and six kinds of alkaloids were identified from carbonized Nelumbinis Folium.After processing,nuciferin,N-nornuciferine,anolobin and neferine that existed in the fresh Nelumbinis Folium disappeared from the carbonized one and the chromatographic peak areas of the other six kinds of alkaloids reduced significantly.CONCLUSION Significant changes occurred in alkaloids composition of carbonized Nelumbinis Folium.Three unknown components appaered in the carbonized one and should be investigated further.
