摘 要:从雄性不育相关基因克隆入手,开展红麻雄性不育机理的研究。根据不同物种atp1基因的保守序列设计引物,在红麻质核互作型雄性不育保持系中通过RACE克隆得到atp1基因cDNA全长,全长为2315 bp,和其他物种相比较,其同源性大于94%。该研究为下一步构建表达载体进行转基因表达研究提供了基础,为揭示红麻雄性不育的分子机理提供了一定依据。Gene cloning was conducted to study the mechanism of male sterility in kenaf (Hibiscus cannabinus). According to the conserved sequence of gene atpl in different species, primers were de- signed. Full length of cDNA of gene atpl were cloned from the cytoplasmic - nuclear male sterile maintai- ner line by using the method of RACE. The gene was 2315 bp in length. This gene shares more than 94% identity of the sequence compared with other species. This study would provide the foundation for studying transgenic express of atpl by constructing expression vector, which will supply certain basis to reveal the mechanism of male sterility in kenaf.
