摘 要:建立了一种简单、快速、准确地测定地肤中齐墩果酸质量分数的高效液相色谱法,以V(甲醇)∶V(水)=9∶1为流动相,检测波长210 nm,柱温为室温,流速1 mL/min,进样量10μL。在上述色谱条件下,齐墩果酸保留时间为11.55 min,精密度及稳定性相对标准偏差分别为2.14%和2.28%,回收率为92.89%。应用响应面法优化齐墩果酸提取条件,其最优提取条件为提取温度65℃,提取时间30 min,m(地肤种子)∶V(甲醇)=1 g∶20mL,并在此基础上比较了地肤不同部位中的齐墩果酸质量分数。结果表明地肤种子中齐墩果酸质量分数最高,达到(41.18±2.02)mg/g,其次是根,为(30.21±1.22)mg/g,叶、茎中质量分数最低。A simple, rapid, sensitive and synchronous method for determining the oleanic acid in Kochia scoparia was developed. The chromatographic conditions are: methanol/water (9/1 v/v) of the mobile phase, 210 nm of detection wavelength, 1 mL/min of flow rate and 10 μL of inject volume. The retention time of oleanic acid is 11.5 min, the relative standard deviation of precision and stability are 2.14% and 2.28%, respectively, and the recovery is 92.89%. The ultra- sonically assistant extraction conditions of oleanic acid from K. scoparia are optimized by using response surface methodology, and the optimal extraction conditions are : 65℃ of extraction temperature, 30 min of extraction time and 1 g : 20 mL of the solid-water ratio. Compared with the oleanolic acid content in different parts of/E scoparia, the oleanic acid content is the highest in the seeds, 41.18±2.02 mg/g, followed by the roots, 30.21±1.22 mg/g, and the lowest in leaves and stems.
