摘 要:目的测定缺血再灌注损伤模型肾组织中肾损伤分子-1(KIM.1)表达,探讨其在反映肾脏热缺血损伤方面的应用价值。方法建立肾缺血再灌注损伤模型,根据热缺血时间长短随机将大鼠分为对照组(热缺血0min组)、热缺血15min组、热缺血30min组、热缺血45min组和热缺血60min组。再灌注8h后,苏木素-伊红(HE)染色观察肾脏病理变化,检测血清肌酐(Cr)值,免疫组织化学法检测肾组织KIM-1表达、Westernblot法检测胞质中KIM.1的含量。结果随着热缺血时间的延长,肾小管肿胀、坏死程度加重,血清cr值增高,由(32.1±5.7)i.Lmol/L升高至(96.8±9.5)txmol/L,肾组织KIM-1表达增强,累积吸光度(M)值由3724-62升高至40928±238。45min热缺血时间内,肾组织KIM.1表达与Cr值呈正相关(r=0.91,P〈0.05)。结论大鼠肾组织KIM-1随热缺血时间延长而表达增强,可联合病理学检查,从形态学和功能学两方面反映热缺血损伤对肾脏功能的影响程度。Objective To verify weather kidney injury molecular-1 ( KIM-1 ) could reflect the function of rat kidney undergoing warm ischemia injury (WII). Methods An ischemia reperfusion model was established. Based on the length of warm isehemia time, rats were divided into control group ( no WII), 15-min WII group, 30-min WII group, 45-min WlI group and 60-min WII group. After reperfusion for 8 h, allograft samples were collected and cut into sections for hematoxylin and eosin (HE) staining, immunohistochemistry and Western blotting, respectively. Blood was collected for further tests. Results With the extension of the warm isehemia time, the kidney tubules became swollen and necrotic. Cr went from (32. 1 ± 5.7 ) μmol/L to (96. 8 ± 9. 5 ) μmol/L, hut cumulative absorbanee value (1A) of KIM-1 went from 372 ± 62 to 40 928 ± 238. The KIM-1 expression was correlated with Cr (r = 0. 91, P 〈 0. 01 ). Conclusion As a biomarker, the expression of KIM-1 get stronger and stranger with the the extension of warm isehemia time. In combination with pathological examination, KIM-1 could reflect function of the kid- ney under WIL
