摘  要:对怀山药 ( Dioscorea opposita)零余子愈伤组织的诱导、分化、再生苗的生根和移栽进行了研究。结果表明 :( 1 )在不同激素组合的培养基上怀山药零余子均能产生愈伤组织 ,而且具有一次成苗的能力。BA2 mg/L+ NAA2 mg/L的培养基对诱导愈伤组织最有利 ,其出愈率达 1 0 0 % ;( 2 )在愈伤组织的分化中 ,BA1 mg/L+ NAA1 mg/L的激素组合是最佳的 ,其分化率为 63.6% ,且多形成丛生芽 ;( 3)再生植株生根和壮苗的最好激素组合为 KT2 mg/L+NAA0 .0 2 mg/L+ PP3330 .1 mg/L,生根率为 91 .2 % ,且根系发达 ,叶色浓绿 ,生长健壮。再生植株移栽后生长健壮 ,对环境具有较好的适应性。This paper deals with studies on \%in vitro\% culture of bulbil of Dioscorea opposita ,including callus induction and differentiation,plantlet root development and transplantation.Results showed:(1) Calli were obtained from bulbil on media containing different kinds of auxin and cytokinin,also the plantlets could be developed at the same time.It was advantageous to callus induction on MS basal medium supplemented with BA 2 mg/L and NAA 2 mg/L.The frequency of callus induction was 100%;(2) The combination of BA 1 mg/L and NAA 1 mg/L was the best to callus differentiation of which ratio was 63.6%.Most of calli formed multiple buds;(3) 91.2% of the regenerated plantlets treated with KT 2 mg/L+NAA 0.02 mg/L+PP333 0.1 mg/L could develop their roots,grew robustly and robusted green leaves;after the regenerated plantlets were transplanted to soil,they grew robustly and robusted green leaves,moreover could adapt to enviroment.
