摘 要:研究了去污剂十六烷基三甲基溴化铵(CTAB)和β-环糊精(β-CD)联合作用(人工伴侣系统),对变性—还原溶菌酶的复性作用。考察了操作条件对溶菌酶复性收率的影响。结果表明,在人工伴侣系统辅助下,60min可使变性溶菌酶的复性收率达89%,自发复性仅为27%;并且其最佳复性温度为37℃,最佳pH值为7~9;同时发现,单独使用环糊精也可在一定程度上辅助溶菌酶复性,复性收率为46%。This article deals with the refolding of denatured-reduced lysozyme promoted by artificial chaperones, that is, cetyl trimethyl ammonium bromide (CTAB) and (β-cyclodextrin(β-CD). The effects of operation conditions on the refolding yiled were investigated. The refolding yiled can rise to 89% in a hour assisted by of artificial chaperones. Under the same condition, there is 27% recovery of lysozyme activity spontaneously. Moreover, the optimal temperature (i.e. 37℃) and pH (i.e.7 to 9) for artificial chaperones-facilitated lysozyme refolding operation were determined. Although lysozyme may be refolding to some extent (46%) only assisted by β-CD.
