摘 要:目的 观察兔脊髓缺血再灌注损害时β-七叶皂甙的保护作用,并探讨其可能的作用机制。方法 家兔12只,随机分为缺血再灌注组(组Ⅰ,n=6)和β-七叶皂甙组(组Ⅱ,n=6)。肾下阻断腹主动脉40min后开放,组Ⅱ于阻断前10min,静脉注射β-七叶皂甙1mg/kg后,继以0.15mg/kg·h微量泵持续给药至主动脉开放;组Ⅰ则以同样方法静脉注射等容量生理盐水作对照。采不同时相动脉血,分别测定血浆血管紧张素-Ⅱ(AT-Ⅱ)和丙二醛(MDA)水平。术后观察脊髓神经功能和脊髓形态学变化。结果 β-七叶皂甙明显降低脊髓缺血期间AT-Ⅱ和MDA浓度,改善术后神经功能,减轻脊髓病理学损害。结论 β-七叶皂甙对脊髓缺血性损伤有良好保护作用,其机制可能与抑制AT-Ⅱ的活性和抗过氧化反应有关。Objective To observe the protective effects of β-Aescin on spinal cord ischemia in rabbits and explore its mechanisms.Methods 12 rabbits were randomly divided into ischemia group(group I )and β-Aescin group (group Ⅱ ) . The in-frarenal aortic was released following clampin for 40 minutes. In group Ⅱ , β- Aescin(1mg/kg) was intravenously given 10 min before clamping and continued at a rate of 0.15 mg/kg.h until unclamping. Normal saline was used as control in group I . Plasma AT -Ⅱ, MDA were measured at different intervel during the experiment. The post-operation neurologic outcomes were evaluated and the morphology of spinal cord were observed. Results β-Aescin induced a decrease in the levels of plasma AT-Ⅱ and MDA during spinal cord ischemia and improved neurologic outcomes. Conclusion β-Aescin can protect the spinal cord from ischemia injury. The mechanisms maybe related to inhibiting over-release of oxygen free radical and AT - Ⅱ .
