欧洲鳗鲡种苗使用孔雀石绿后的残留评估

摘  要:将初始体质量为(5.0±0.5)g的欧洲鳗鲡,经质量浓度为0.2mg/L孔雀石绿浸浴24h后换水1/2,再以0.1mg/L孔雀石绿浸浴24h,经2次换水,每次换水2/3,将污染鱼体捞出,移入无污染的水泥池,另以相同规格未经孔雀石绿浸浴的欧洲鳗鲡作为对照组,按《无公害欧洲鳗鲡静养池塘养殖技术规范》进行正常养殖管理。定期抽取试验组和对照组鳗鲡各10尾,测定规格后,采用高效液相色谱-串联质谱联用仪测定鳗鲡体内的孔雀石绿及无色孔雀石绿残留,孔雀石绿和无色孔雀石绿的方法检测限均为0.5μg/kg。结果表明,试验组鳗鲡养殖至240d,鱼体体质量达100g时,鳗鲡体内未检出孔雀石绿,而无色孔雀石绿残留量仍达(2.2±0.4)μg/kg;养殖至300d,鱼体体质量达182g时,无色孔雀石绿未检出;对照组试验过程均未检出孔雀石绿和无色孔雀石绿残留。孔雀石绿及无色孔雀石绿在欧洲鳗鲡体内代谢缓慢,且无色孔雀石绿在鳗鲡体内残留时间较孔雀石绿更长,种苗期间使用孔雀石绿,极易导致孔雀石绿在鳗鲡养成品中残留。The European the concentration of 0.2 eel seedlings at an average mg/L bath water for 24 h, weight of(5 _+ 0.5 ) g were exposed to malachite green (MG) at and then half of the water was changed. The seedlings were ex- posed to malachite green (MG) at the concentration of 0. lmg/L bath water for another 24 h, during which two - thirds of the water was changed twice. After treatment, the contaminated eels were transferred into a cement pool and treated as the test group. Another group of eels without MG treatment was taken as the control group. Both groups were cultivated in normal ways. At regular time, 10 eels were extracted respectively from both groups for LC/MS/MS analysis. The detection limit of malachite green(MG) and leuco - malachite green(LMG) were both 0.5p^g/kg. The results indicated that MG residues in European eels was not detected up to 240 d , when the eels were at the weight of 100 g ,and 300 d was needed for LMG, when the eels were at the weight of 182 g. The control group had no MG and LMG residues in the test process. MG and LMG both eliminated very slowly from European eels and LMG eliminated slower than that of MG. It was found that the use of malachite green in seedling green residues in eels when cultivated to adult fishes.
