摘 要:目的观察腺相关病毒载体9型(AAV9)介导Zs Green基因在体转染心肌组织的效果。方法采用冠状动脉灌注法将携带报告基因Zs Green的腺相关病毒9型(AAV9-Zs Green)以不同滴度(5×106TU,1×107TU)转导至大鼠心肌组织,分别于转染后1、2和4周在荧光显微镜下观察大鼠心肌组织绿色荧光表达情况。结果病毒载体转染1周后心肌组织可见绿色荧光表达;2周时可见大量绿色荧光表达,荧光强度较1周时明显增强(P<0.01);4周时荧光强度较2周有所减弱(P<0.05),但仍有较强表达;随着病毒转导滴度提高,荧光强度明显增加(P<0.05)。结论腺相关病毒载体能够有效介导Zs Green转染大鼠心肌组织,并长期稳定表达。【Objective】 To investigate the efficiency of Adeno-associated virus9(AAV9) vector-mediated Zs Green gene delivery to rat myocardial tissue in vivo.【Methods】 Various titers of virus with Zs Green gene were delivered to the heart of SD rats through coronary infusion.The expression of green fluorescence in myocardial tissue was examined at 1,2 and 4 weeks after transduction.【Results】 The expression of green fluorescence was observed 1 week after transduction,and boosted in 2 weeks(P 0.05).Though the fluorescence intensity was still strong in 4 weeks,the fluorescence intensity was reduced compared with that in 2 weeks(P 0.05).Fluorescence intensity of the high titer group was significantly higher than the low titer group(P 0.05).【Conclusion】 AAV vector with Zs Green gene was successfully transfected into the rat heart,and the expression of gene was long term and stable.
