摘  要:【目的】通过对陆地棉(Gossypium hirsutum L.)PHD(Plant homeodomain)转录因子在棉花不同组织和非生物胁迫条件下的表达分析,筛选棉花逆境胁迫应答基因。【方法】根据已报道的28个棉花GhPHD基因的EST序列合成了特异引物,使用半定量RT-PCR方法检测GhPHD在棉花种子、根、茎、叶、纤维(15DPA)中的表达以及在干旱、高盐(150 mmol/L NaCl)、低温(4℃)非生物胁迫下的表达。【结果】棉花PHD转录因子在各组织中的表达存在差异,85.7%的基因在种子中表达,在纤维中表达的占35.7%。在干旱胁迫条件下GhPHD1、GhPHD6和GhPHD22呈上调表达,在高盐胁迫条件下GhPHD1、GhPHD16呈上调表达,低温胁迫激活GhPHD9表达,其它基因对三种非生物胁迫无明显应答。【结论】部分GhPHDs在陆地棉应答和适应非生物胁迫过程中可能发挥重要的作用。[ Objective] In order to identify novel genes involved in upland cotton (Gossypium hirsutum L. ), plant stress adaptation, expression of GhPHDs under different kinds of abiotic stresses, such as drought, high salt and low temperature were analyzed. [ Method ] According to the reported 28 GhPHDs, specific primers were synthesized. Semi - quantitative RT - PCR method was used to analyze expression patterns of GhPHDs in cotton seed, root, stem, leaf and fiber(15DPA) and under abiotic stresses such as drought, high salt (150 mmol/L NaCI) and low temperature (4~C). [ Result] The 28 GhPHDs showed different tissue - specific expression patterns in seed, root, stem, leaf and fiber(15DPA) of cotton. The proportion of genes expressed in seed was 85.7% and 35.7% in fiber. These genes were differentially regulated by various abiotic stresses. The expressions of GhPHD1, GhPHD6 and GhPHD22 were activated by drought stress, and GhPHD1, GhPHD16 were up - regulated by high salt stress. GhPHD9 was up - regulated by low temperature. Other genes didnt show obvious response to these three abiotic stresses. [ Conclusion] It implies that GhPHDs may play important roles during upland cotton plant adapted to abiotic stresses.
