摘 要:目的探讨川芎嗪对过氧化氢诱导大鼠离体培养晶状体氧化损伤的抗氧化作用。方法 SD大鼠24只,随机分8组,即正常对照组、过氧化氢对照组、不同浓度川芎嗪组(0.125 mg/ml、0.25 mg/ml、0.5 mg/ml、1 mg/ml、2 mg/ml和4 mg/ml)。摘除大鼠眼球,取晶状体进行体外培养,24 h后取出晶状体,肉眼观察晶状体混浊程度,然后匀浆取上清液,通过黄嘌呤氧化酶反应法检测超氧化物歧化酶(SOD)活性;通过丙二醛可与硫代巴比妥酸缩合法检测丙二醛(MDA)含量。结果 SOD检测结果显示,与过氧化氢组比较,0.5 mg/ml、1 mg/ml和2 mg/ml的川芎嗪浓度可明显升高SOD活性,P<0.05或P<0.01。MDA检测结果显示,与过氧化氢组比较,各个浓度的川芎嗪均可降低晶状体MDA的含量,P<0.01。结论川芎嗪可抵抗过氧化氢所致的大鼠晶状体的氧化损伤,其机制可能与提高SOD活性和降低MDA含量有关。Objective To observe the in vitro antioxldative effect of tetramethylpyrazine (TMP) on hydrogen per- oxide induced oxidative damage of mt lens. Method Twenty-four SD rats were randomly assigned into 8 groups: normal control, hydrogen peroxide treated, and hydrogen peroxide treated plus different concentration of tetmmethylpyrazine (0. 125 mg/ml, 0.25 mg/ml, 0.5 mg/ml, 1 mg/ml, 2 mg/ml and 4 mg/ml). The concentration of hydregen peroxide (H2O2) that used to produce oxidative damage was 800μmol/L. The lenses were enucleated and cultured under the differ- ent conditions for 24 h. The opacity of the lenses was observed by naked eyes and the cultural supernatants were isolated. The levels of active supemxide dismutase (SOD) and nudon aldehyde (MDA) were measured by photometry method re- spectively. Result The active SOD levels significantly higher in groups that were treated with 0.5 mg/m1,1.0 mg/m1 or 2.0 mg/ml TMP ( P 〈0.05 orP 〈0.01). The MDA content was significantly lower in all groups that were treated with TMP ( P 〈 0.01 ). Conclusion Tetramethylpyrazine can protect rat lenses from oxidative damage, as demonstrated by the increased active SOD level and decreased MDA content in the present study.
