VitC对高糖下视网膜毛细血管周细胞增殖的影响

摘　　要：目的　观察 Vit C对高糖下体外培养视网膜毛细血管周细胞增殖的影响 ,探讨糖尿病视网膜病变 (DR)的防治方法。方法　以新鲜牛眼球为材料 ,分离并培养周细胞 ;采用3H- Td R掺入法及液体闪烁技术 ,研究不同浓度的 Vit C(5 0 ,1 0 0 ,5 0 0 ,1 0 0 0 ,1 5 0 0 mg/ L)对周细胞增殖的影响。结果　 5 0 0 ,1 0 0 0 mg/ L的 Vit C对生理浓度葡萄糖 (5 mmol/ L)下周细胞的增殖没有影响 ,大剂量 Vit C(5 0 0 ,1 0 0 0 ,1 5 0 0 mg/ L)加入高糖 (2 0 mmol/ L)培养液中培养周细胞 48h,对周细胞的增殖有明显促进作用。结论　大剂量 Vit C可促进高糖下周细胞的增殖 ,提示 Vit C可作为临床防治Objective To observe the effect of VitC to retinal capillary pericytes proliferetion under high glucose and research the prevention and treatment of diabetic retinopathy(DR).Method Retinas were dissected from eyes of fresh killed cattle,separated and cultured retinal capillary pericytes.The effect of VitC(50,100,500,1000,1500mg/L)to pericytes'proliferetion was evaluated by 3H thymidine incorporation and liquid scintillation techniques.Result There was no significant difference in VitC(500,1000mg/L)to the proliferation under normal glucose (5mmol/L),but VitC of high dose(500,1000,1500mg/L)could promote the proliferation under high glucose(20mmol/L)culturing for 48h.Conclusion VitC of high dose could promote pericytes' proliferation under high glucose,so it could be of interest to prevent and cure DR clinically.
