摘  要:目的探讨重组人生长激素(rhGH)对THP-1单核细胞TNF-α和IL-6分泌的影响,并研究其与NF-κB活性的相关性。方法应用ELISA检测rhGH诱导THP-1细胞培养上清中TNF-α和IL-6的分泌情况,并观察LPS和NF-κB特异性抑制剂BAY11-7082对其分泌的影响;应用荧光素酶报告基因和电泳迁移率实验(EMSA)检测NF-κB在rhGH诱导的单核细胞中的活化程度。结果 rhGH单独可以促进THP-1细胞分泌TNF-α和IL-6,抑制LPS诱导的细胞因子的分泌;BAY11-7082能显著抑制rhGH和LPS诱导的TNF-α和IL-6的分泌,NF-κB活性与TNF-α和IL-6的分泌呈现明显的相关性。结论 rhGH可通过NF-κB信号通路双向调节THP-1单核细胞TNF-α和IL-6的分泌。Objective To investigate the effect of recombinant human growth hormone (rhGH) on the secretion of TNF- α and IL-6 in THP-1 ceils and analyze the correlation between the secretion and NF-κB signaling pathway. Methods The concentrations of TNF-α and IL-6 in the supernatants of THP-1 induced by rhGH were measured with ELISA. The effects of LPS and an NF-KB inhibitor, BAYll-7082 on the secretion of TNF-α and IL-6 were also observed. The activity of NF-KB was detected by luciferase reporter and electrophoretic mobility shift assay (EMSA) in THP-1 cells induced by rhGH. Results rhGH alone promoted the secretion of TNF-α and IL-6 in THP-1 cells while inhibited the secretion of TNF-α and IL-6 in THP- 1 stimulated by LPS. BAYlI-7082 inhibited the secretion of TNF-α and IL-6 in THP-1 stimulated by LPS and rhGH. The activity of NF-κB was significantly correlated with the secretion of TNF-α and IL-6. Conclusion rhGH played a bidirectional role in the regulation on the secretion of TNF-α and IL-6 via NF-κB signaling pathway in THP-I cells.
