中药益气通络丹含药血清对大鼠血管平滑肌细胞周期的影响

摘　　要：目的 :观察益气通络丹 (YQTLD)含药血清对培养SD大鼠血管平滑肌细胞 (VSMC)细胞周期的影响 ,探讨该药抗冠心病、脑血栓等心脑血管疾病的作用机制。方法 :用血清药理学方法制备含药血清 ,用流式细胞术 (FCM )测定作用2 4h后含药血清对VSMC细胞周期的影响情况。结果 :益气通络丹含药血清能使SD大鼠VSMC的G1期细胞所占百分数增加 ,细胞增殖率 (PI)值降低 ,与空白血清对照组有显著性差异 (P <0 .0 1)。结论 :益气通络丹含药血清能阻滞细胞停留在G0G1期 ,抑制细胞进入S期 ,有抗VSMC的增殖作用 ,这可能是其临床上抗冠心病、脑血栓的作用机制之一。Objective: To study effect of serum containing YQTLD on cell cycle of SD rat's culture VSMC.Method: Serum containing YQTLD was prepared by serum pharmacological method. VSMC cultured was treated with the serum. The effect of the medicine on cell cycle of VSMC was assayed by flow cytometery.Results: The results showed serum containing YQTLD could make the percent of G0G1 phase increase remarkably and the value of PI decrease remarkably( P < 0.01 ) comparing with the control serum.Conclusion: It suggested that the mechanism might be relevant with the influence of inhibition proliferation on VSMC by blocking the cell stay at G0G1phase, prevent it entering S phase.
