摘 要:目的观察胰高糖素样肽-1(GLP-1)类似物利拉鲁肽(LRL)对高糖及脂多糖(LPS)诱导的人脐静脉内皮细胞(HUVECs)核转录因子-κB(NF-κB)和单核细胞趋化蛋白1(MCP-1)表达的影响。方法体外培养HUVECs,细胞传至第3代,随机分为7组:正常对照组(C组),高糖组(HG组),LPS组,高糖+LPS组(HGS组),高糖+LRL组(HGL组),LPS+LRL组(LPSL组)和高糖+LPS+LRL组(HGSL组)。CCK-8法检测LRL干预前后HUVECs的增殖情况,RT-PCR和ELISA分别检测NF-κB、MCP-1 mRNA和蛋白的表达情况。结果与C组比较,HG组、HGS组、LPS组HUVECs的生长受到明显的抑制(P<0.05),LRL干预后各组细胞的生长抑制情况得到减轻(P<0.05)。HG组、LPS组、HGS组HUVECs中NF-κB、MCP-1的mRNA和蛋白水平较C组升高(P<0.05);LRL干预后各组mRNA和蛋白水平降低(P<0.05)。结论高糖和LPS抑制HUVECs增殖,促进NF-κB、MCP-1表达增加,LRL能有效降低炎症因子的表达,改善内皮功能紊乱,有助于延缓糖尿病动脉粥样硬化的进程。【Objective】To investigate effects of liraglutide(LRL) on the expressions of nuclear factor-kappaB(NF-κB) and monocyte chemoattractant protein-1(MCP-1) mRNA and protein in human umbilical vein endothelial cells(HUVECs) cultured under high glucose and LPS environment,and its mechanisms of anti-diabetic artherosclerosis.【Methods】HUVECs were cultured in vitro and at the 3rd generation were divided into seven groups: normal control group(group C),high glucose group(group HG),LPS group(group LPS),HG plus versus concentration of LPS group(group HGS),group HG plus LRL(group HGL),group LPS plus LRL(group LPSL),group HGS plus LRL(group HGSL).CCK-8 used to detect the proliferation of HUVECs in different groups before and after LRL intervention;mRNA expressions of NF-Κb,MCP-1 were determined by RT-PCR;the protein of NF-κB,MCP-1 were determined by ELISA.【Results】The proliferation of HUVECs in HG,LPS and HGS groups were lower than C group(P 0.05),after LRL treatment they became higher than the latter(P 0.05).The mRNA and protein expression levels of NF-κB and MCP-1 in HUVECs of HG,HGS and LPS groups were higher than that of C group(P 0.05).The mRNA and protein expression levels of NF-κB and MCP-1 in cells treated with LRL were lower than before treatment(P 0.05).【Conclusion】LRL plays a key role in delaying atherosclerosis by decreasing the expression of NF-κB and MCP-1.
