摘 要:目的探讨肾康注射液对大鼠肾缺血再灌注损伤的影响及其机制。方法将36只SD大鼠随机分为随机分为假手术组(S组)、缺血再灌注组(I/R组)和肾康注射液治疗组(T组),双肾缺血60 min后恢复灌注,术后24 h检测血清尿素氮(BUN)和肌酐(Scr),观察肾组织超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量以及肿瘤坏死因子(TNF)-α、白介素(IL)-1β和IL-10水平。结果与S组相比,I/R组BUN和Scr升高、肾组织SOD活力下降、MDA含量、TNF-α、IL-1β和IL-10水平均升高;而与I/R组相比,T组BUN([37.74±4.39)比(48.56±4.75)mmol/L]和Scr([197.25±17.43)比(248.63±19.25)μmol/L]降低,肾组织SOD活力([69.95±5.43)比(60.46±3.18)u/(mg.prot)]和IL-10水平([63.59±2.49)比(56.37±1.64)ng/L]上调,MDA含量([9.28±2.70)比(14.71±3.46)nmol/(mg.prot)]、TNF-α([169.72±5.67)比(182.64±6.84)ng/L]和IL-1β([10.62±0.69)比(13.53±0.94)ng/L]水平下调。结论肾康注射液可以减轻大鼠肾缺血再灌注损伤,其机制与减轻氧化应激、抑制炎症反应有关。[ Objective ] To investigate the protective effects and its mechanism of Shenkang Injection against rats with renal isehemic reperfusion injury. [ Methods ] Rats were randomly divided into sham operation group (group S), isehemic reperfusion group (group I/R) and Shenkang Injection group (group T), 12 rats each. Model of rats was established by both kidneys reperfusion followed 60 min duration of renal ischemia. Twenty-four hours after operation, blood urea nitrogen and serum creatinine were detected, superoxide dismutase activity, malondialdehyde contentrations and levels of tumor necrosis factor-or, interleukin-l and interleukin-10 of renal tissue were determined. [ Results ] Compared with those in the group S, blood urea nitrogen, serum creatinine, malondialdehyde concentrations and levels of tumor necrosis factor interleukin-l and interleukin-10 of renal tissue were increased and superoxide dismutase activity of renal tissue were decreased in group I/R. Blood urea nitrogen [(37.74±4.39) vs. (48.56±4.75) mmol/L], serum creatinine [(197.25±17.43) vs. (248.63 ±19.25) umol/L], malondialdehyde contentrations [(9.28±2.70) vs. (14.71±3.46) nmol/mg'prot] and levels of tumor necrosis factor-a [(169.72±5.67) vs. (182.64±6.84) ng/L] and interleukin-ll3 [(10.62±0.69) vs. (13.53±0.94) ng/L] of renal tissue were lower in group T than in group I/R, superoxide dismutase activity [(69.95±5.43) vs. (60.46±3.18)u/(mg "prot)] and level of interleukin-10 [(63.59±2.49) vs. (56.37±1.64) ng/L] of renal tissue were higher in group T than in group I/R. [Conclusion] Shenkang Injection can ameliorate with renal ischemic reperfusion injury in rats, partly through attenuation of oxidative stress inhibition of inflammatory reaction.
