摘 要:目的探讨Egr-1(early growth response-1)蛋白、MMP-2、TGF-β1在肾小管间质纤维化大鼠模型中的表达及意义。方法采用单侧输尿管结扎术(Unilateral Ureteral Obstruction,UUO)致肾小管间质纤维化大鼠模型。将54只SD大鼠随机分为假手术组、模型组、贝那普利(Benazepril)治疗组[10 mg/(kg.d)]。手术后第5、10和15天分别处死各组中6只大鼠,用免疫组化方法观察各组大鼠肾小管间质中Egr-1蛋白,转化生长因子-β1(transforming growth factor-β1,TGF-β1),基质金属蛋白酶-2(matrix metalloProteinase-2,MMP-2)表达情况。行HE、Masson染色后进行图像分析评定各组肾小管间质损害程度。结果模型组各时间点肾小管间质损伤指数及Egr-1蛋白、TGF-β1表达明显高于假手术组(P<0.05),MMP-2表达明显低于同期假手术组(P<0.05);而贝那普利组各时间点肾小管间质损伤指数及Egr-1蛋白、TGF-β1表达明显低于模型组(P<0.05),MMP-2表达明显高于同期模型组(P<0.05)。结论 Egr-1蛋白参与肾小管间质纤维化过程;贝那普利在肾小管间质纤维化中对肾脏有保护作用。[Objective] To investigate the expression and significance of Egr-1 protein,MMP-2 and TGF-[31 in tubnlointerstitial fibrosis. [ Methods ] 54 normal female Sprague-Dawley rats were randomly divided into 3 groups: Sham-operated controls, unilateral ureteral obstruction (UUO), benazepril treatment [10 mg/(kg'd)]. 6 rats of each group were sacrificed randomly on the days 5, 10, 15 after surgery. Immunohistochemistry was performed on ob- structed renal tissue to detect early growth response-1 (Egr-1), transforming growth factor-J3~ (TGF-~l) and matrix metallo-proteinase-2(MMP-2). Histological studies were also done by HE and Masson staining. [Results] The tubu- lointerstitial damage indexes, the expression of Egr-1 protein and TGF-[31 in UUO group at different time points were significantly higher than in sham-operated group (P 〈0.05), the expression of MMP-2 was significantly lower than in sham-operated group (P 〈0.05); the tubulointerstitial damage indexes, the expression of Egr-1 protein and TGF-~I in benazepril group at different time points were significantly lower than in UUO group(P 〈0.05), the expression of MMP- 2 was significantly higher than in UUO group (P 〈0.05). [Conclusion] Egr-1 protein plays a very important role in mediating the tubulointerstitial fibrosis; Benazepril has protective effect on tubulointerstitial fibrosis.
