中药联合推按运经仪对家兔胆囊CCK、VIP受体基因表达的影响

摘　　要：目的:观察中药胆胃舒冲剂联合推按运经仪对胆囊收缩素(CCK)和血管活性肠肽(VIP)受体基因表达的影响及预防胆囊结石的作用。方法:新西兰大耳白兔45只,随机分为3组,每组15只,空白组喂养普通饲料;模型组喂养普通饲料加诱石饲料;治疗组喂养同模型组,并同时喂胆胃舒冲剂,并加行推按运经仪治疗。实验8周后观察3组胆囊结石情况及胆囊壁CCK、VIP受体基因表达。结果:治疗组胆囊结石形成率、CCK受体基因表达、VIP受体基因表达等指标与模型组比较,差异均有显著性意义(P<0.05)。结论:胆胃舒冲剂联合推按运经仪可降低胆囊结石的形成,提高胆囊CCK受体基因表达,同时降低VIP受体基因表达,从而加强胆囊收缩功能,促使胆汁排泄,防止胆囊结石的发生。Objective： To observe the effect of Chinese medicine Danweishu Granules combined with massage instrument on the gene expression of cholecystokinin receptor（CCK-AR） and vasoactive intestinal peptide receptor（VIP-R） in rabbit gallbladder, and to explore its preventive mechanism for gallstone Methods： Forty-five New Zealand rabbits were equallyrandomized into 3 groups. The blank control group was fed with normal diet, model group was fed with nomal diet and lithogenic diet, and treatment group was fed with nomal diet, lithogenic diet and Danweishu Granules. Meanwhile, the treatment group was given treatment of massage instrument. After feeding for 8 weeks, the animals were sacrificed and sampled, and the expression of CCK-AR and VlP-AR in gallbladder tissue was examined by RT-PCR assay. Results： Compared with the model group, increase of gallbladder contraction rate and the expression of CCK-AR, and the decrease of VIP-R were found in the treatment group （P 〈0.05）. Conclusion： Chinese medicine Danweishu Granules combined with massage instrument can prevent the occurrence of gallstone, and its therapeutic mechanism is possibly related with the increase of CCK-AR gene expression and with the decrease of VIP-AR gene expression.
