摘  要:目的观察健脾益肾化瘀通络方对链脲佐菌素(STZ)诱导的Wistar大鼠糖尿病肾病(DN)肾脏保护作用及对氧化应激的影响。方法 74只Wistar雄性大鼠,按45 mg/kg体重给予大鼠空腹一次性腹腔注射STZ进行造模。模型成功后,按体质量随机分为正常组、模型组、厄贝沙坦组(17.5 mg/kg)、健脾益肾化瘀通络方小剂量(30.8 mg/kg)和大剂量(123.2 mg/kg)组。每4周测体质量、24 h尿蛋白定量;14周时处死大鼠,记录肾重与体重比值;检测血清甘油三酯(TG)、尿素氮(BUN)、肌酐(Scr)、一氧化氮(NO)、丙二醛(MDA)和超氧化物歧化酶(SOD)浓度;进行肾组织HE、Mallory、PASMΡ-HE染色,观察肾组织病理变化并进行肾组织病理评分分析。结果与模型组比较,健脾益肾化瘀通络方不同剂量组体质量显著增加(P<0.05),肾重体重比明显降低,可以明显改善肾组织病理变化;血清TG、BUN、Scr明显改善(P<0.05),血清SOD活性升高(P<0.05),同时MDA含量减少(P<0.05)。结论健脾益肾化瘀通络方可能通过拮抗DN大鼠的氧化应激作用而对肾脏起到有效地保护作用。Objective To observe the kidney protection effect and influence of Jianpi Yishen Huayu Tongluo Fang on oxidative stress in rats with diabetic nephropathy (DN) induced by streptozocin (STZ). Methods Male Wistar rats (n = 74 ) were intraperitoneally injected STZ (45 mg/kg) for establishing model, and then divided, after successful modeling and according to body weight, into normal grouop, model group, irbesartan (17.5 mg/kg) group and low-dose (30. 8 mg/kg) and high-dose (123.2 mg/kg) Jianpi Yishen Huayu Tongluo Fang groups (low-dose group and high-dose group). The body weight and 24-hour urine protein quantitate were measured every 4 weeks. On the 14th week the rats were executed for recording the ration of kidney weight to body weight (KW/BW) and detecting the levels of serum triglyceride (TG), blood urea nitrogen (BUN), serum creatinine (Ser), nitric oxide (NO), malondialdehyde (MDA) and superoxide dismutase (SOD). The pathological changes and scores of kidney tissue were observed and analyzed after staining of HE, group and high-dose group, the body weight increased significantly, pathological changes of kidney tissue were Mallory and PASM-HE. Results In low-dose significantly ( P 〈 0. 05 ), KW/BW decreased relieved, levels of serum TG, BUN and Scrdecreased significantly ( P 〈 0. 05 ), activity of serum SOD increased ( P 〈 0. 05 ) (P 〈 0. 05 ) compared with model group. Conclusion Jianpi Yishen Huayu Tongluo of protecting kidney through antagonizing oxidative stress in DN rats. and MDA decreased Fang can play a role
