摘  要:目的探讨加味补阳还五汤对糖尿病(DM)大鼠视网膜微血管周细胞凋亡的影响。方法采用链脲佐菌素诱导DM大鼠模型,将DM大鼠分为治疗组(给予加味补阳还五汤10.71g/kg)、对照组(羟本硫酸钙120 mg/kg)、空白组(生理盐水3 mL/d),并设立正常组对照。造模成功后连续给药12周,12周后取大鼠眼球制备视网膜血管消化铺片,行PAS染色,观察视网膜微血管形态,测量单位视野内微血管网面积率,计数单位视野内周细胞总数,并采用TUNEL法检测视网膜微血管周细胞的凋亡。结果 4组大鼠视网膜微血管排列规则,毛细血管管径粗细均匀,单位视野内微血管网面积率和周细胞总数4组间差异无统计学意义(P>0.05)。正常组大鼠未见视网膜微血管周细胞凋亡,DM大鼠在第12周已经出现视网膜微血管周细胞的凋亡,治疗组与正常组比较差异无统计学意义(P>0.05),治疗组较对照组、空白组凋亡程度轻,差异具有统计学意义(P<0.05)。结论 DM大鼠在第12周已经出现视网膜微血管周细胞凋亡,加味补阳还五汤能抑制DM大鼠视网膜微血管周细胞的凋亡。Objective To investigate the influence of Jiawei Buyang Huanwu Tang on apoptosis of retinal microvessel pericytes in rats with diabetes mellitus (DM). Methods The rat model of DM was induced with streptozotoci, and DM rats were divided into treatment group treated with Jiawei Buyang Huanwu Tang (10.71 g/kg), control group given calcium dobesilate (120 mg/kg) and blank group given normal saline(3 mL/d), and additionally a normal group was set up. After successful modeling, the rats were given medicinal for 12 weeks. Retinal digest preparations were prepared and given PAS taining for observing the morphology of retinal microvessels. The area rate of microvessel net per sight was detected and the total pericyte count per sight was calculated. The apoptosis of retinal microvessel pericytes was detected by using terminal-deoxynucleoitidyl transferase mediated nick end labeling (TUNEL). Results The arrange of retinal microvessels was regular, diameters of capillaries were uniform, and differences in area rate of microvessel net and total pericyte count per sight had no statistical significance among 4 groups (P 〉 0. 05 ). There was no apoptosis of retinal microvessel pericytes observed in normal group, but there was in DM rats on the 12th week. The comparison between treatment group and normal group had no statistical significance ( P 〉 0. 05 ), and the apoptosis group and blank group ( P 〈 0. 05 ). Conclusion appeared in DM rats on the 12th week, and Jiawei was milder in treatment group compared with control The apoptosis of retinal microvessel pericytes has Buyang Huanwu Tang can inhibit the apoptosis.
