摘  要:目的:探讨二氧化硅(SiO2)刺激的矽肺患者肺泡巨噬细胞(AM)培养上清中IL-1β的表达及其协同其它因子对人胚肺成纤维细胞(FB)增殖的作用。方法:收集矽肺患者肺灌洗液中的AM,在体外以SiO2(50μg/ml)和不加血清的DMEM培养3、6、12、18、24、36小时,采用ELISA法检测不同时间点AM上清中IL-1β的表达,并用MTT和3H-TdR法检测AM培养上清作用于FB的细胞增殖情况。结果:经SiO2刺激的矽肺患者AM上清中IL-1β的表达明显上调,与对照组同时间相比,差异具有显著性(P<0.01),IL-1β表达在体外培养12小时达最高峰;AM培养上清各时间点均有促进FB增殖的作用,18小时上清促增殖作用最强,且经SiO2刺激的AM上清促细胞增殖作用更显著(P<0.05,P<0.01)。结论:SiO2再刺激的矽肺患者AM培养上清可以促进人胚肺FB的增殖,其机制之一可能为上调AM中IL-1β的表达进而影响细胞因子网络调控。Objective:To investigate the expression of IL-1β which had been provoked by SiO2in alveolar macrophages supernatant, and the role combined with other factors on the proliferation of human lung fibroblasts. Methods : Human alveolar macrophages were collected by bronchoalveolar lavage of silicosis patient, cultured AMs in non-serum culture medium with or without silicon dioxide on 3, 6, 12, 18, 24, 36 hour and detected the expression of IL-1β in the AMs supernatant with enzyme linked-immuno-sorbent assay. Then proliferation of human lung fibroblasts by the AMs superuatant was measured with MTT and 3 H-Thymidine methods. Results:The expression of IL-1β in the AMs supernatant which had been exposed to SiO2 increased significantly with prolonged time, compared with the control group (P 〈 0.01 ) . Its expression arrived the maximum at 12 hour. The patient AM supernatant could accelerate proliferation of lung fibroblasts. The superuatant of AM culture for 18 hours had the most strong effect. It was more obvious in AM supernatants exposed to SiO2 (P 〈 0.05, P 〈 0.01 ). Conclusion : Through AM mediation SiO2 can accelerate proliferation of lung fibroblasts, the mechanism of one may be raised expression of IL-1β in the AMs, then influence cytokine network regulation.
