金纳米粒子分光光度法测定尿中8-羟基脱氧鸟苷

摘  要:在pH为3.5的BR缓冲溶液中,8-羟基脱氧鸟苷(8-OHdG)与金纳米粒子作用,导致溶液吸光度比值改变,且在0.498×10-6~12.7×10-6mol/L范围内,吸光度值改变与8-OHdG的浓度呈良好的线性关系,其回归方程为:ΔA630/A520=0.081 2c-0.007 13,相关系数r=0.998 4,检出限为0.150×10-6 mol/L,相对标准偏差(RSD)在1.48%~3.57%。用于尿中8-OHdG的测定,回收率在96.1%~98.0%。该方法简单、快速、成本低。In the Britton-Robinson buffer solution of pH 3.5,8-hydroxy-2 ' -deoxyguanosine (8-OHdG) combined with gold nanoparticles, resulting in absorbance change of the solution. A linear correlation existed between the absorbance change and the concentration of 8-OHdG in the range of 0. 498 × 10-6 - 12.7 ×10 -6 mol/L. The equation of linear regression is △A630/A520 = 0.081 2c -0.007 13 with a correlation coefficient (r) of 0. 998 4. The detection limit (LOD) is 0. 150 × 10-6 mol/L. The relative standard deviations were 1.48% - 3.57%, and the recovery was 96.1% - 98.0%. The proposed method was simple, rapid and cheap.
