摘 要:目的探讨内毒素性急性肺损伤(ALI)过程中P38丝裂原活化蛋白激酶(P38 MAPK)的表达。方法腹腔注射脂多糖(LPS)20mg/kg建立小鼠ALI模型。心脏穿刺查血气分析,并获取肺脏标本,观察肺组织形态学改变。采用免疫组织化学法和Western blot技术检测小鼠肺组织中磷酸化P38MAPK的表达。结果与对照组比较,ALI组各时相点小鼠肺组织中磷酸化P38MAPK阳性表达明显增加,且在2h达到峰值,主要分布于浸润的炎症细胞、上皮细胞、血管内皮细胞。结论内毒素性ALI过程中启动了P38MAPK信号传导通路。Objective To investigate the expression of P38 MAPK in endotoxin-induced acute lung injury(ALI). Methods The ALI model was set up by intraperitoneal injection of 20 mg/kg lipopoiysaccharide(LPS). Blood samples were collected for blood gas analysis,lung samples were for histomorphology changes. The expressions of phosphated p38 MAPK in lung tissues were detected by immunohistochemistry and Western blot. Results The positive expressions of phosphated P38 MAPK distribute mainly in infil trative inflammatory cells, epithelial cells and endothelial cells. And the levels of expression of phosphated P38 MAPK in ALl group were higher obviously than those in control group,peeking at two hour. Conclusion P38 MAPK signal transduction pathway was activated in endotoxin-induced ALI.
