摘 要:目的观察表没食子儿茶素没食子酸脂(Epigallocatechin Gallate,EGCG)对中脑原代培养细胞的作用。方法采用大鼠胚胎中脑原代细胞培养法,培养中脑神经细胞,再将细胞分为空白对照组、阳性对照组、实验组,给予EGCG和神经生长因子(nerve growth factor,NGF)进行配组干预,然后观察细胞生长状况,同时运用四唑盐(MTT)比色法检测神经细胞活性;用抗酪氨酸羟化酶(tyrosine hydroxylase,TH)对多巴胺能神经元(dopaminergic neurons,DA)行免疫荧光染色,比较各组阳性神经元数目及突起的变化情况。结果实验组(EGCG组)培养细胞生长旺盛,轴突延长,相互交错。MTT法测得细胞吸光度值(A值)及免疫细胞化学TH阳性细胞数实验组与空白对照组比较差异有统计学意义(P<0.05),实验组与阳性对照组(NGF组)比较二者差异均无统计学意义(P>0.05)。结论 EGCG对中脑原代培养细胞及DA神经元有一定的保护作用。Objective To investigate the effects of Epigallocatechin Gallate(EGCG)on the primarily cultured neuronal cells from embryonic rat mesencephalon. Methods Primarily cultured neurons were prepared from embryonic rat mesencephalon after treated with EGCG and NGF in different deliveries in vitro. Mesencephalic cells were observed and their viability was assessed by MTT assay and the number of tyrosine hydroxylase (TH) positive cells was counted with immunofluorescence. Eventually, the number of positive cells and their neurites of outgrowth were compared between different groups. Results The number of TH-positive cells and the viability of mesencephalic cells were significantly higher in EGCG groups or positive control groups (NGF groups) than in the control groups (P 〈 0.05). But there was not markedly difference between the experimental groups and the NGF groups ( P 〉 0.05). Conclusions The results suggest that EGCG may have some protective effect on mesencephalic cultured cells from rat embryos.
