摘 要:合成了准刚性环化合物2,3-二氢-9,10-二羟基-1,4-蒽醌(DHHA),并用核磁共振谱、红外光谱和质谱确定了结构.循环伏安实验表明DHHA在玻碳电极上有一对明显的氧化还原峰,在加入DNA后,其氧化峰电位出现了正移,且峰电流出现了明显的降低,表明二者的作用模式可能为沟槽结合,当DNA浓度处于9.0×10–6mol/L-4.5×10–5mol/L之间时,DNA的浓度与氧化峰的电流强度具有良好的线性关系,所以,在此范围内可以对DNA进行定量检测.计时库仑实验、微分脉冲实验以及紫外-可见光谱实验都进一步证实DHHA与DNA的相互作用,且它们的作用模式可能为沟槽结合.The quasi-rigid ring compound 2,3-dihydrogen-9,10-dihydroxy-1,4- anthraquinone (DHHA) was synthesized. Then the structure was determined by IHNMR, IR and MS spectrum analytical methods. Cyclic voltammetry experiment revealed that DHHA had a pair of obvious redox peak at the glassy carbon electrode. After interaction with DNA, the redox peak currents underwent significant decreased and peak potentials showed positive shift tendency, indicating that DHHA might bind with DNA through a groove binding mode. At the same time, it showed that the concentration of DNA could be qualified in the range of 9.0×10-6mol/L-4.5×10-5mol/L via using DHHA as an electroactive probe. In addition, chronocoulometry, differential pulse voltammetry and UV-Vis spectrophotometry methods further confirmed that the interaction between DHHA and DNA, and the action mode of them might be combined with groove.
